Objective:This study is exploring the expression of the proteins and mRNAs of kallikrein-related peptidase6(KLK6) in normal cervical tissue, moderate-severe cervical intraepithelial neoplasia, cervical carcinoma and then investigating the relationship with clinical pathological features of cervical carcinoma.Method:1. The immunohistochemical staining streptavidin-preosidase method was applied to detect the expression of KLK6protein in10normal cervical samples,15CINII-Ⅲ samples,77cervical carcinoma samples;and60of them (10normal cervical samples,10CINII-Ⅲ samples,40cervical carcinoma samples) were fresh tissues, we use the real-time fluorescent quantitative PCR technology to detect the expression of KLK6mRNA, then analyzing the correlation the expression of KLK6in cervical carcinoma.2. Analyzing the protein and the mRNA expression of KLK6in cervical carcinoma.3. Analyzing the relationship between the expression of KLK6protein and mRNA in the same with the clinical and pathological factors of cervical carcinoma. Analyzing the consistency of KLK6protein and mRNA expression in the40fresh cervical cancer cases4. Analyzing the relationship between the expression of KLK6protein with the clinical and pathological factors of cervical carcinoma.5. Analyzing the relationship between the KLK6protein expression level and pelvic lymph node metastasis in cervical carcinoma.The data was analyzed with the statistics software SPSS17.0and set inspection level a at0.05.Results:1. The positive expression rates of KLK6protein in normal cervical, CINII-Ⅲ and cervical carcinoma were10.0%,40.0%,83.1%. The differences between the three groups have statistics significance. The positive expression rates of KLK6protein in cervical carcinoma were significantly higher than that in CINII-Ⅲ and normal cervical tissue, there were statistical significance. The strong positive expression rates of KLK6protein in cervical carcinoma were significantly higher than that in CINII-Ⅲ tissue, there were statistical significance.2. The expression levels of KLK6mRNA in normal cervical tissue, CINII-Ⅲ and cervical carcinoma were1.172±0.68、2.950±1.366.14.378±9.652. The difference between the three groups have statics significance (P=0.000<0.05).3. KLK6protein and mRNA of the40fresh cervical cancer cases positively correlated.4. The expression of KLK6protein in cervical carcinoma was significantly associated with clinical stage, histological differentiation degrees, pelvic lymph node metastasis, myometrium invasion depth (P<0.05), however age, histological type, high risk HPV infection,vascular invasion depth and tumor diameter did not show strong association(P>0.05).5. The single factor comparative analysis between cervical carcinoma pelvic lymph node metastasis group and no cervical carcinoma pelvic lymph node metastasis group demonstrates the expression of KLK6protein, were associated with cervical carcinoma pelvic lymph node metastasis (p<0.05), and further logistic regression analysis shows that KLK6protein level histological differentiation degrees, muscle invasion depth were independent risk factors of cervical carcinoma pelvic lymph node metastasis (p<0.05).Conclusion:1. The expressions of KLK6protein and mRNA in cervical carcinoma tissue were significantly higher than that in CINII-Ⅲ and normal cervical tissue. The expressions of KLK6protein and mRNA in cervical carcinoma were positively correlated, which suggested that KLK6may take part in the process of the cervical carcinoma.2. The expression of KLK6protein in cervical carcinoma were associated with clinical stage, histological differentiation degrees, pelvic lymph node metastasis, myometrium invasion depth, which suggested that KLK6protein was associated with cervical carcinoma invasion and metastasis.3. KLK6protein expression level is an independent risk factor of cervical carcinoma pelvic lymph node metastasis.
