The Effect Of Astaxanthin On Sepsis-induced Hyperglycemia And Serum Adiponectin

Objective:Through cecal ligation and puncture to establish the rats model of sepsis to study astaxanthin activation on sepsis in rats survival rate, hyperglycemia, serum adiponectin and organ protection.Methods:Septic rats model was made by standard cecal ligation and puncture (CLP).(1) Forty eight rats were randomly divided into4groups(n=12):sham group、CLP group、CLP+vehicle group (CV), CLP+Asta group (CA), observed within7days survival rate of each group.(2)Thirty two rats were randomly divided into4groups(n=8) Sham+vehicle group (SV), Sham+Asta group (SA), CV group, CA group, sham group animals underwent anesthesia and laparotomy only. CLP group animals received operation of CLP. Blood samples were collected at preoperative, postoperative,6h,12h,24h from caudal vein for fast blood sugar.24h after operation, intraperitoneal glucose tolerance test was implemented. Detect insulin levels and blood glucose at30min,60min,120min. Collect blood sample for detect the12h and48h serum adiponectin。The optical microscopy was used to observe pathological morphologic change of heart, liver, lung, kidney tissue which was gotten48hours after surgery.Results:(1) CLP group and CV group within7days survival rate was33.33%, significantly lower than control group100%and CA group75%.(2) Blood glucose of post-operation was higher in every group than pre-operation(P<0.01);6h,12h,24h after operation, CV group blood sugar was higher than the other three group(P<0.05).(3) After intraperitoneal glucose injection, CV group blood glucose at30min,60min,120min were higher than in CA group; serum insulin level was lower in CV group than in CA group at each time point(P<0.05).(3) Serum adiponectin level at48h in each group was lower than at12h, CA group was higher than CV group at12h and48h (P<0.05).(4) From H-E staining we observed that the inflammation cells infiltration in CA group was much gently than in CV group about heart, liver, lung, kidney tissue, and its structure is relatively complete.Conclusion:Astaxanthin can increasing serum adiponectin and insulin concentration thus can improve hyperglycemia in the rats model of sepsis, also on cardiovascular, liver, lung, kidney have certain protective effects and can improve the survival rate of sepsis in rats.
