Background and Aims: Interleukin 17D(IL-17D)is involved in the pathogenesis of various diseases.As an important immune cytokine,its role in sepsis is still unclear.This topic mainly explores the role of IL-17 D in sepsis and further studies its mechanism of action.Methods: The concentration of IL-17 D in serum of patients with sepsis and healthy subjects was detected by ELISA.The mouse sepsis model was established by cecal ligation puncture(CLP)to detect IL-17 D protein concentration in mice by ELISA.Intraperitoneal injection of IL-17 D protein after mice underwent CLP,survival rate,cell sorting and counting,bacterial load,liver and kidney functions and HE stained sections were observed.cytokine concentration levels were detected after treatment of IL-17 D.Phagocytosis and bacterial killing function of macrophages and bone marrow neutrophils wered observed after IL-17 D treatment,then signaling pathways were screened.Results: The IL-17 D protein concentration in patients with sepsis was raised.The concentration of IL-17 D was elevated in blood and peritoneallavage fluid(PLF)in mice after CLP.after Intraperitoneal injection of IL-17 D protein to CLP mice,the survival rate was significantly lower than that of the control group,the bacterial load was increased,the liver and kidney function and HE staining sections showed more serious tissue damage.after IL-17 D treatment of primary macrophages,macrophage phagocytosis was decreased and NF-?B phosphorylation was decreased.Conclusions: IL-17 D is involved in the immunopathological process of sepsis and affects the phagocytosis function of macrophages by inhibiting the phosphorylation of NF-?B signaling pathway which could influence the bacterial clearance ability of mice,thus aggravate the onset of sepsis.
