Experimental Research On Jianpi Xiaozheng Fang For Human Liver Cancer HepG2Cell Apoptosis And Its Mechanism

Objective:The purpose of this study was to investigate:①The inhibiting effect of Jianpi Xiaozheng Fang in vitro on the proliferation of human Liver cancer cell line HepG2、SMMC-7721;②The effect of Jianpi Xiaozheng Fang on the apoptosis of HepG2;③The apoptosis mechanism of Jianpi Xiaozheng Fang treatment on the HepG2.Method:①MTT method was used to observe the effects of Jianpi Xiaozheng Fang on human Liver cancer cell line HepG2, SMMC-7721proliferation;②AnnexinV/PI double staining method was used to observe the early apoptosis induced by different concentrations of Jianpi Xiaozheng Fang in human Liver cancer cell line HepG2;③Real-time PCR method to detect apoptosis-gene expression after Jianpi Xiaozheng Fang treatment Liver cancer cell line HepG2.④Western Blotting method to detect apoptosis-protein expression after Jianpi Xiaozheng Fang treatment on Liver cancer cell line HepG2.Results:①Jianpi Xiaozheng Fang inhibited the proliferation of human Liver cancer cell line HepG2SMMC-7721.②Jianpi Xiaozheng Fang treatment on human Liver cancer cell line HepG2for48h,induced the apoptosis of HepG2with a certain degree of concentration-dependence.③Jianpi Xiaozheng Fang treatment on human Liver cancer cell HepG2,Cells appeared apoptosis characteristic form.④Jianpi Xiaozheng Fang treatment on human Liver cancer cell HepG2,Cell ultrastructure morphological changes in apoptosis.⑤Jianpi Xiaozheng Fang treatment on human Liver cancer cell HepG2for48h, the lever of ROS was rised.⑥Jianpi Xiaozheng Fang treatment on Liver cancer cell HepG2down-regulate apoptosis-gene Bcl-2, Survivin and up-regulate apoptosis-gene Bak,Bax.⑦Jianpi Xiaozheng Fang treatment on Liver cancer cell HepG2, up-regulate apoptosis-pretein Bax, caspase-3, caspase-9, down-regulate apoptosis-protein Bcl-2.Conclusions:①Jianpi Xiaozheng Fang inhibited the proliferation of human Liver cancer cell HepG2, SMMC-7721.②Jianpi Xiaozheng Fang can induce human Liver cancer cell HepG2apoptosis.③Jianpi Xiaozheng Fang has an effect of inducing apoptosis,The possible mechanisms might be down-regulate apoptosis-gene Bcl-2, Survivin,up-regulate apoptosis-gene Bak,Bax and up-regulate apoptosis-pretein Bax, caspase-3, caspase-9, down-regulate apoptosis-protein Bcl-2.
