Purpose:To explore the correlation between the expressing rules andthe spinal cord functional recovery after spinal injury in rats with RGMB(repulsive guidance molecule B).Methods:50adult male SD rats, divided into two groups randomly,25of which degraded without the application of chondroitin sulfate(Chondroitinase ABC, ChABC)(group A); the other25degraded with theapplication of chondroitin sulfate (Chondroitinase ABC, ChABC)(group B)After the operation,the two groups will be observed and recorded thesituation of their body movements and executed for test specimen at thesame time respectively. BBB score method is used to evaluate the hindlegs feeling and the movement function of the tested rats. HE stain is tocalculate the whole area of the damage part; Real-time PCR is to detectRGMB mRNA expression; the application of Immunofluores cence stainingis to test the double standard expression of collagen fibre acidicprotein/neurofilament protein-200(neurofilament-200, NF-200) Results:The BBB score of group B was obviously higher than that ofgroup A2to4weeks after injury at the same period (P<0.05); Two weeksafter the injury,The HE staining result showed that hollow formed in eachgroup of damaged area,and damage zone area in group A is significantlyless than group B (P<0.05). GFAP/NF-200double immunofluorescencestaining GFAP expression gradually lower in group B2to4weeks, and theFluorescence intensity values of Group A is significantly higher than that ofGroup B. It is also higher than group A (P<0.05) with NF-200immunohistochemical staining positive fibers going through the damagedarea; RGMB mRNA expression is significantly declined compared withGroup A (P<0.05). The difference was statistically significant.Conclusions:It is negatively correlated with the spinal cord functionalrecovery after spinal cord injury in rats RGMB concentration in themicroenvironment, RGMB inhibitors may be helpful for clinical treatmentof spinal cord injury.
