Objective:In this study,NSCs will be seeded into the directional PLGA scaffolds and transplanted into the hemisected thoracic spinal cord of adult rat.The effect of PLGA seeded with NSCs in the repairing of the hemisected spinal cord of rat and the MRI of the model rat will be investigated.Methods:NSCs from the spinal cord of the embryonic rat were cultured in vitro.The PLGA which NSCs were seeded into was implanted into thorax8-9hemisected SCI rat.The rats were divided into3groups randomly:only SCI group,NGF-PLGA group,NSCs+NGF-PLGA group.Behavioral analysis was performed on all rats using BBB open field locomotion scale every two weeks after operation.The rats were examined by7.0T MRI to identify the axonal regeneration at12hours,12weeks and24weeks after operation.The spinal cord tissuses of NSCs+NGF-PLGA group rats were observed with Immunohistochemistry to identify the survival,migration,differentiation of the grafted cells.Results:The BBB score of PLGA group rats and NSCs+NGF-PLGA group rats were higher than the score of only SCI group (P<0.05).There was no evidence of MRI to identify the axonal regeneration.In the NSCs+NGF-PLGA group rats,the neural stem cells were survival and differentiate into the neural cells.Conclusions:Transplanting neural stem cells within PLGA scaffolds has contribution to function recovery of SCI rats.The MRI is a useful examination to identify the neural axon regeneration.
