Objective:Through the detection of eight loci candidate genes HLA-DP rs3077, rs2301220, rs3135021, rs9277341, rs9277378, rs9277542, rs9277535, rs9277471single-base polymorphism to investigate Xinjiang uygur and han nationality HLA-DPA1, HLA-DPB1SNPs and their correlation with chronic hepatitis b; Compared with xinjiang uygur in patients with hepatitis b and its national health in HLA-DPA1, HLA DPB1-the difference between SNPs. Compared with Xinjiang uygur and han population and the han nationality in patients with chronic hepatitis b patients with chronic hepatitis b HLA DPA1, Gene polymorphism of HLA-B1and HLA-DPA1, B1single nucleotide polymorphisms and xinjiang Uighur population in occurrence and development of chronic hepatitis b. Contrast the Uighur hepatitis b patients and their healthy crowd this nation in HLA-DPA1, The difference between B1single nucleotide polymorphisms.Methods:To collect inpatients and outpatients from Xinjiang medical university affiliated hospital of traditional Chinese medicine and Korla uygur hospital as subjuects. Received samples are divided into Uighur and Han nationality patients with chronic hepatitis b; Uighurs and Han nationality healthy subjects, our group with eaual number. Adopted method is the case-control study. The cases are the chronic hepatits b patients, while the control group is the halthy medical staff group.(1) Applying the DNA extraction kit to conduct the genomic DNA extraction specimens and quality inspection (2) Useing the sNapshot method for HLA-DP gene rs3077, rs2301220, rs3135021, rs9277341, rs9277378, rs9277542, rs9277535, rs9277471eight sites for detection and classification.3, Using SPSSS17.0, SAS software, statistical analysis, etc., In the experiment data, aga can be shouwn by mean and standard deviation,and genotype frequency and allele frequency can be described by proportion or absolute number are Hardy differences-and you Weinberg dotted with x2test and t test, P<0.05, explain differences is significant, with statistical significance.Results:(1) rs3O77in HBV+compared to healthy controls (χ2=8.323, P=0.0156), rs2301220(x2=9.470, P=0.0088), rs9277341(χ2=11.923, P=0.0026) for the three sites for P values are less than0.05, statistically significant, so its locus polymorphism is associated with HBV infection.(2)Compared with controls, the han nationality HBV+rs3077(χ2=9.313, P=0.0095), rs2301220(χ2=9.566, P=0.0084), rs9277341(χ=13.096, P=0.0014) three loci P values are less than0.05, with statistical significance.(3)In healthy controls the han and uygur healthy controls, rs3077(χ2=28.096, P<0.0001, rs2301220(χ2=21.637, P <0.0001), rs9277341(χ2=24.467, P<0.0001), rs9277378(χ2=16.695, P=0.0002), rs9277471(χ2=14.946, P=0.0006), rs9277542(χ=16.074, P=16.074), rs9277535(χ2=18.645, P<0.0001) in the seven sites of single-base parting with statistical significance.(4)The han nationality HBV+ and Uighur HBV+ comparison, rs3077(χ2=45.675, P<0.0001), rs2301220(χ2=38.934, P<0.0001), rs9277341(χ2=47.344, P<0.0001), rs9277378(χ2=26.661, P<0.0001), rs9277542(χ2=24.309, P<0.0001), rs9277471(χ2=29.360, P<0.0001) of the six bits click single-base parting with statistical significance.(5)In the research of gender and hepatitis b virus infection were found in healthy controls against male vs female rs9277341(χ2=9.271, P=0.0097), rs9277378,(χ2=7.936, P=0.0189), rs9277542(χ=8.364, P=0.0153), rs9277471(χ2==8.194, P=0.0166) four loci of single-base parting is statistically significant.ConcIusions:(1)HLA-DP gene rs3077, rs2301220, rs9277341, rs9277378, rs9277542, rs9277535, rs9277471seven bits of some different allele number of A, G, C, T and gene frequency of significant differences in uygur and han nationality, HLA-DP genotype of single-base points are ethnic differences. While rs3135021two alleles of A, the number and frequency of the G no obvious difference between the two peoples.(2)HLA-DP SNP rs3077, rs2301220, three sites of rs9277341classification associated with hepatitis b virus infection, to illustrate the three sites may be associated with hepatitis b virus infection.(3)The Uighur hepatitis b patients and healthy controls, found eight loci associated with hepatitis b virus infection had no statistical significance, this may be the Uighur prevalence of hepatitis b because of lower.(4)Compared the healthy han and uygur group, rs3077, rs2301220, rs9277341, rs9277378, rs9277542, rs9277471significant differences these six genetic loci, shows that the single nucleotide polymorphisms of the differences between the two peoples, it is probably causes of Uighur hepatitis b is lower than Han Chinese hair.(5)Gender differences and the relationship between hepatitis b virus (HBV) infection, rs9277341, rs9277378, rs9277542, rs9277471four single nucleotide polymorphism loci previously may be related to the proportion of the male than female.
