Objective To investigate the expression and significance of lactoferrin in human gliomas and research the effect of lactoferrin at differernt concentration affects VEGF and bFGF in C6cell line in vitro.Methods We collected46cases of primary human glioma in the neurosurgery department of XiangYa hospital. The gene and protein expression of lactoferrin were detected by immunohistochemical, RT-PCR and Western-Blotting methods in10case normal brain tissues and46case of human glioma that contains27case of grade Ⅰ~Ⅱ and19case of grade Ⅲ~Ⅳ.To observe the effect of lactoferrin at different concentration on VEGF and bFGF.Lactoferrin at concentration of0.006,0.013,0.025,0.050g/L,and black control groups were included in this research. Results The gene and protein expression of lactoferrin is different in normal brain tissue and glioma,even in different grade in glioma.Lactoferrin expresses in grade Ⅰ~Ⅱ glioma was lower than nomal brain tissues,and there is no positive expression or obvious decrease in grade Ⅲ-Ⅳ.The positive expression rate of lactoferrin in grade Ⅲ-Ⅳ glioma was significant lower than that in normal brain tissues and grade Ⅰ~Ⅱ gliomas.The RT-PCR and werstern blotting assay proved that VEGF and bFGF mRNA and protein was on the decline while the lactoferrin concentration increased.It showed that lactoferrin inhibits the expression of VEGF and bFGF in the C6cell lines.Conclusion The expression of lactoferrin was different in normal brain tissues and different grade gliomas.It revealed that lactoferrin influences the occurrence and development of glioma.The hign malignancy of high grade glioma maybe related with the lack of lactoferrin.Lactoferrin at different concentration influences the expression of VEGF and bFGF in C6cell lines in vitro.The expression of gene and protein of VEGF and bFGF were on the decline while the concentration of lactoferrin increased.It revealed that lactoferrin can inhibit the glioma by inhibiting the expression of VEGF and bFGF.
