Objective: Cirrhosis is an irreversible fibrosis of the liver caused by various internal and external factors.The incidence and mortality of liver cirrhosis are high in the world.Because early cirrhosis usually has no typical clinical symptoms,it does not often attract the attention of patients.It is often in the middle and late stage of liver cirrhosis,and its manifestation is portal hypertension and hypofunction,and even some cases such as digestive tract hemorrhage,hepatorenal syndrome,and so on.Primary liver cancer and other serious complications.Therefore,the pathogenesis and treatment of liver cirrhosis are hot.However,studies have shown that vascular endothelial growth factor vascular endothel Ial growth factor,VEGF,bFGF basic fibroblast growth factor and Endostatin are involved in angiogenesis,tissue fibrosis and endothelial cell proliferation.Methods:1 Clinical data collection: from January 2015 to February 2018,48 patients with liver cirrhosis treated in Department of Hepatobiliary surgery,second Hospital of Hebei Medical University,were selected as experimental group and 30 normal liver tissues as normal group.2 Experimental methods: HE and Masson trichrome staining were used to observe and compare the changes of liver tissue structure between the experimental group and the control group.At the same time,immunohistochemistry and ELISA were used to detect the difference of expression of VEGF,bFGF and Endostatin protein between the experimental group and the normal group,and the statistical analysis was made.3 Data analysis: All the data are analyzed by SPSS 21.0 software.The data of measurement data are expressed by the standard deviation of mean soil.The measurement data that accord with normal variance are measured by One-Way ANOVAA,one factor analysis of variance.SNK method was usedto compare the two steps.Non-parametric test was used to measure the statistical data which did not accord with normal or uneven variance.There was statistical significance with P<0.05.The statistical chart drawing was carried out by WPS software.Results:1.The results of HE staining and Masson trichrome staining showed that,compared with the normal group,the number of fibrous bands in liver tissue of the experimental group was obviously increased,the pseudolobules were formed,inflammatory cells were widely distributed,and the structure of the catchment area disappeared obviously.Hepatocyte necrosis in varying degrees,fiber proliferation is more obvious;2.Immunohistochemistry: The result of relative gray value analysis shows:The AO of VEGF,bFGF and Endostatin in normal group were0.06±0.02,0.08±0.01 and 0.07±0.02,respectively.The AO of VEGF,bFGF and Endostatin in the experimental group were 0.16±0.02,0.14±0.03 and0.14±0.01,respectively.Compared with the normal group,the expression of VEGF,bFGF and Endostatin was up-regulated in the experimental group,and the difference was statistically significant(P<0.05).3.ELISA test: The potency of VEGF,bFGF and Endostatin in normal group were 280.06±12.22,30.06±2.41 and 4340.52±643.56(pg/ml),respectively.The potency of VEGF,b FGF and Endostatin in the experimental group were 530.35±158.25,63.48±15.38 and 8180.75±1310.34(pg/ml),respectively.The expre-ssion of VEGF,bFGF and Endostatin were significantly up-regulated in the experimental group as compared with the normal group,and the differences were statistically significant(P<0.05).Conclusions: The expression of VEGF,b FGF and Endostatin protein were significantly up-regulated in patients with liver cirrhosis,and the expression of these proteins was closely related to the occurrence and development of cirrhosis.In the pathophysiological process of liver cirrhosis,vascular proliferation and tissue structure change are related to the expression of VEGF and bFGF.The expression of these two proteins may regulate theprogression of cirrhotic diseases.The physiological function of Endostatin protein may also inhibit the progression of liver cirrhosis.The expression of Endostatin may be associated with some regulatory factors in the occurrence and development of cirrhosis or cirrhosis.In a positive feedback role.
