Objective:To investigate the expression of bone morphogenetic proteins15(BMP-15) in the damaged ovaries and serum, whether it can react to the ovarian reserve function.Methods:20adult SD rats with normal estrous cycle were randomly divided into two groups:the blank matched control group, cyclophosphamide (CTX) group. Two groups of rats were subjected to intraperitoneal injection of a single-dose cyclophosphamide (75ml/kg) and saline, respectively. Increasing degree of rat weight and change of ovary, numbers of the follicle and histomorphological features of ovary were observed. ELISA (Enzyme-linked immunosorbant assay) was used to analyze serum level of FSH (Follicle-stimulating hormone), E2(Estradiol)and BMP-15among the two groups. The expression of BMP-15was evaluated by immunohistochemistry, and BMP-15mRNA expression determined by Real-Time PCR in rat ovaries. Results:1. The average ovarian weight and the increasing degrees of the rat weight of CTX group were lower than that of the control group (P<0.05).2. FSH level of CTX group was higher than the control group, E2and BMP-15level of CTX group were lower than the control group (P<0.05).3. Serum levels of BMP-15was positively correlated with E2(P<0.05).4. As compared with the control group, CTX treatments produced reduction in the number of primordial follicle, preantral follicle, antral follicle and the total follicle (P<0.05). But the reduction of second follicle number was not significant (P>0.05).5. The expression of BMP-15in the ovaries of CTX group was lower than the control group (P<0.05).6. As compared with the control group, the expression of BMP-15mRNA in the CTX group was lower (P<0.05).Conclusion:1. Cyclophosphamide can cause obvious ovarian damage.2. CTX can cause a lower expression of BMP-15.3. BMP-15can response to ovarian reserve function of cyclophosphamide-induced rats.
