Background:The transcription regulartory factor-Idl is named as inhibitor of DNA binding or inhibitor of differentiation. It is a kind of trscription regulatory factor with double helix ring structure (helix-loop-helix, HLH). There are four kinds of Ids was found in its family up to now. They are Id1, Id2, Id3and Id4. Id1paticipates the regulator process of cell growth, cell development, cell mature and cell death. The constitutive gene expression of Idl not only promotes cell proliferation, but also inhibits cell differentiation. So the over expression of Idl in cells is closely related with cell malignant growth. It has been reported that Id1was over expressed in several kinds of tumors, and its over expression was related with the degree of cell malignant.Objective:To detect the Id1mRNA by real time PCR and the expression of Idl protein by immunohistochenmistry in human benign and malignant ovarian tumors, and to analysis the relationgship between Idl expression and the malignant or prognosis of ovarian cancer. To identify the clinical diagnosis significance of Idl detection in the gene and protein level.Methods:The relative quantity of Id1mRNA was detected by real time RT-PCR in human benign and malignant ovarian tumor samples. The relationship between the quantity of Idl mRNA and some malignant parameters, such as benign or malignant change, cell differatiation and prognosis, was further analyzed. The protein expression of Idl was futher detected by immuohistochemistry method in the samples of ovarian endometrioid carcinoma and benign chocolate cyst of ovary. The relationship between the expression of Id1and the malignant of ovarian cancers was further analyzed.Results:The mean quantity of Idl mRNA in ovarian cancers was obviously higher than that in ovarian benign samples (P<0.01). Over expression of Idl mRNA was correlated with the pathological type, and was negatively correlated with the differentiation level of ovarian cancer (P<0.01). The level of Idl mRNA was closly correlated with the prognosis (P<0.01). No positive expression of Idl protein was observed in normal epithelial ovarian samples. Few positive expression of Idl protrin was observed in benign chocolate cyst of ovary. All samples of ovarian endometrioid carcinoma showed positive expression of Id1protein. There was higher expression of Id1protein in low differentiation samples than that in moderate or high differentiation samples (P﹤0.01)Conclusion:The expression of Id1in ovarian cancers was obviously higher than that in ovarian benign samples. The over expression of Idl is obviously correlated with the malignant degree and prognosis in ovarian cancer. Detection of Idl mRNA by real time PCR and protein by immonohistochenmistry can help the early diagnosis and prognosis of ovarian cancer.
