The Experimental Research Of Bi Xie Rheumatism-relieved Prescription On High Uric Acid Model For Serum Uric And Uric Acid Salt Transport Protein OAT1and URAT1

Elevated blood uric acid is an important biochemical indicator of gout.However, there is still no long-time safe, effective and economic drug for high uric acid hematic disease or gouty arthritis,. Traditional Chinese medicine has some curative effect, but on the whole disease-oriented study is insufficient.It is of great significance to strengthen the clinical research of traditional Chinese medicine prescriptions to develop TCM for uric acid reduction and anti-inflammatory. My master’s thesis points four parts:the first part is Chinese and western medicine research progress of high uric acid hematic disease; The second part is Chinese and western medicine research progress of gout; The third part is related experimental research of Bi Xie rheumatism-relieved prescription treatment for high uric acid mice model; The fourth part is related experimental research of Bi Xie rheumatism-relieved prescription treatment for acute gouty arthritis rats; The fifth part is related experimental research of Bi Xie rheumatism-relieved prescription treatment for high uric acid mice model kidney human urate transportor. Purpose of this study, methods, results and conclusions are reported as follows.Objective:To observe Bi Xie rheumatism-relieved prescription effect for serum interleukin1and interleukin6from gouty arthritis rats. To observe Bi Xie rheumatism-relieved prescription effect for urine transporters URAT1and OAT1in high uric acid mice model.Methods:1、To group kunming male mice at random and to make model by respectively disposable abdominal cavity injecting oxygen oxazine acid potassium and hypoxanthine. To give ALLO(allopurinol) and benzbromarone for control group, and high dose, middle dose and low dose Bi Xie rheumatism-relieved prescriptionrespectively.To observe Bi Xie rheumatism-relieved prescription effect for blood uric acid serum cholesterol and serum triglycerides and to detect urine transporters URAT1and OAT1expression.2、To group SD male mice at random, to make model by crystallization articular cavity injecting uric acid sodium.To give high dose, middle dose and low dose Bi Xie rheumatism-relieved prescription respectively. To give colchicine once per day, seven days in a row. To observe Bi Xie rheumatism-relieved prescription effect in blood IL-1and IL-6.Results:Bi Xie rheumatism-relieved prescription can reduce serum uric acid level from high uric acid hematic model. High dose and middle dose prescription has a certain level of lowering triglycerides. Bi Xie rheumatism-relieved prescription can reduce serum interleukin6and interleukin1from acute gouty arthritis model,which shows that it has certain anti-inflammatory effect. Bi Xie rheumatism-relieved prescription can effectively increase the URAT1expression and down-regulate OAT1expression.Conclusion:1、It is effective and reliable to make high uric acid mice model by methods of intraperitoneal injection and it can be used as a traditional Chinese medicine intervention high uric acid animal model.2、Bi Xie rheumatism-relieved prescription can effectively reduce serum uric acid level from high uric acid hematic model.3、Bi Xie rheumatism-relieved prescription have anti-inflammatory effect and can reduce serum interleukin6and interleukin1from acute gouty arthritis model.4、Bi Xie rheumatism-relieved prescription can effectively adjust the renal uric acid salt transport related proteins and suppress uric acid kidney reabsorption through up-regulating URAT1expression and down-regulating OAT1expression.Research significance:The experiment proves that Bi Xie rheumatism-relieved prescription can effectively reduce serum uric acid、serum triglycerides and have good anti-inflammatory effects. Through the study of renal uric acid salt transport protein expression, we consider that the mechanism of Bi Xie rheumatism-relieved prescription should be related with that. It has great significance to develop new drugs and to study further mechanism.
