| Objective:Discussing possible mechanism of NB-UVB combining psoralea injectionfor the synergistic treatment of vitiligo, throughing research the cell proliferationmelanogenesis,tyrosinase activity and of NB-UVB combining Psoralea injection onA375human melanoma cells.Methods:1.MTT was used to detect the cells number changes by differentconcentration gradient of psoralea injection and different intensity of NB-UVBeffectted on A375cells after cell proliferation curve. Selectting psoralea injection thathad not significant effect on cell profliferation used for subsequent experiments andselecting NB-UVB radiation that had not significant effect on cell profliferation usedfor subsequent experiments.2. Selected the appropriate concentrations of psoraleainjection on A375cells combining with NB-UVB after24h,48h and72-hour,and thenobserved the growth of A375human melanoma cells by MTT, tested the changes oftyrosinase activity with dioxypenylalanine oxidation;detectted the content of melaninwith the law of spallation of NaOH.Results:1.Psoralea injection(concentration0.5%)and irradiated byNB-UVB(22mj/cm2) effectted on A375melanoma cell proliferation:comparing withblank group and intervention groups, cell proliferation was increased obviously at48h and72h(p<0.05) except24h(p>0.05) in the psoralea injection+NB-UVBgroup,and then A490reached maximum was0.924±0.033at72h.2.Psoralea injection(concentration0.5%)and irradiated by NB-UVB(22mj/cm2)effectted on A375melanoma cell melanin synthesis: in the psoraleainjection+NB-UVB group, the melanin content was increased at various degrees,and there were time-dependent relationship, and comparing with blank group andintervention groups, the melanin content reached maximum at72h, and the differenceshowed obvious(P<0.05).3.Psoralea injection(concentration0.5%)and irradiated by NB-UVB(22mj/cm2) effectted on A375melanoma cell tyrosinase activity:comparing with blank group andintervention groups, the psoralea injection+NB-UVB group showed a growingtendency of tyrosinase activity, and there were time-dependent relationship,thetyrosinase activity reached maximum at72h(P<0.05).Conclusion:1.Psoralea injection combining with NB-UVB can promote togetherA375melanocyte proliferation.2. Psoralea injection combining with NB-UVB can increase cell tyrosinase activity,which leads to achieve the effect of promoting melanin synthesis. |
