IS is more common in a clinical three-dimensional spinal deformity, especially occurs inchildren and adolescents, its pathogenesis is complex, and the cause is not yet explicit.Endochondral bone formation is the main way the trunk of bone formation, therefore theupper and lower end of the vertebral endplate cartilage,which is a close relationshipbetween the activity of osteoblasts with scoliosis occurs and subject to a number ofendocrine hormones and signals precise regulation of the system,play a key role in thelongitudinal growth of the spine.In recent years, researchers have confirmed therelationship between Sox9, FGF, TGF and other cytokines and IS formation, however theimpact of the occurrence and development of scoliosis for the Ihh-PTHrP signalingsystem rarely reported. Ihh that is referred to as the Indian porcupine protein and belongsto a subtype of Hedgehog gene family acted a crucial role in process of Endochondralbone formation,and it express the cell hypertrophy and its the first major phase. HumanPTHrP gene mainly by the secretion of cartilage joints that located on chromosome12sand played the role effect of the lower layer cells is the other key factor on proliferation,differentiation and growth of cartilage by inhibition of mast cell apoptosis and furtherdifferentiation of chondrocytes.In this study, it inferred bone cartilage on both sides oftheir differences, further explore the molecular mechanisms of the development ofscoliosis by means of the expression levels of the vertebral endplate of concave side and a convex side at the model of porcine scoliosis.ObjectiveTo explore the expression of Ihh and PTHrP in cartilage and molecular biologymechanism of the porcine scoliosis model, in order to study etiology of human idiopathicscoliosis further, we research the expression level and parts of IHH, PTHrP in swinebilateral scoliosis vertebral cartilage endplate by immunohistochemistry.MethodChoose two2-month-old normal piglets numbered1or2randomly,measured cobbangle and divided into A (convex side), B (center), C (concave side)3groups, each16parts from the cartilage endplate compensatory curve convex side of the vertebral body,the Central and the concave side, in total48copies as the experimental group. Take two2-month-old normal piglets numbered3or4randomly as control group, divided intoD(left side), E(right side) two group by removed from the left side and the right sidecartilage endplate of the curve vertebral body, in total32copies. Detect the expression ofPTHrP and Ihh in5sets of cartilage endplate by immunohistochemistry,Observe the expression level and parts of IHH and PTHrP in each group,and the average optical density was detected in all samples.Results1.HE staining results showed that similar to the growth of long bone plate structure werepresented in each vertebral endplate, it showed pale blue and could be broadly dividedinto the reserve zone, proliferative zone, mature areas and calcification areas, comparedto the convex side of the top vertebrae, the concave side was thinner, mild disordersstructure and less cell number.2.Ihh mainly expressed in the cytoplasm of the proliferating cartilage cells andhypertrophic chondrocytes in cartilage endplate, PTHrP expressed in the cytoplasm ofreserve areas, proliferative zone and maturation zone in cartilage endplate, but mainly expressed in chondrocytes of the proliferative zone and maturation zone.The expressionquantity of Ihh in A, B, C, D, E five groups was0.3504±0.0233,0.2473±0.0286,0.1126±0.0306,0.2189±0.0366,0.2365±0.0226, The expression quantity of PTHrP in A, B, C,D, E five groups were0.4306±0.0154,0.2376±0.0248,0.1473±0.0229,0.2471±0.0218,0.2685±0.0456. Five sets of data was analyzed by one-way ANOVA, results showed thatA group compared to B, C, D, E group, p <0.01, with a significant difference, Bcompared to A, C, D, E,p<0.01, with a significant difference; there was no significantdifference between B, D, E groups, p>0.01.3. The simple correlation analysis was carried out to the expression on the Ihh and PTHrP,result showed that: r=0.585, p<0.01and with statistically significant and positivecorrelation.Conclusion1.The osteogenic activity of convex side in cartilage endplate was significantly higherthan the concave side in the experimental group, while there was no significantdifference between the bilateral in control group.2.The expression of Ihh, PTHrP is A (the convex side)> B (middle)> C (concave side) inthe experimental group, this difference may be related closely to the scoliosis in the pigmodel.3.There has differences of the expression of Ihh and PTHrP in each group and a positivecorrelation is between the expression...
