[Objective] To explore in the genomic level how fascin promotes theoccurrence and progression of tumor through downstream signalingmolecules in the Cholangiocarcinoma cell RBE.[Methods] Using whole-genome Affymetrix microarray to compare thedifference of the gene expression profiles of RBE between Fascin geneRNA interference group and non-interference group and screeningsignificant differentially expressed genes; Using the bioinformaticssoftware to perform Pathway analysis and Gene Ontology analysis of thegenes for initial screening results.[Results] Screening out704genes with significant difference, including466apoptosis genes and238proliferation genes in the interference group,by the comparison of the gene expression profiles of RBE between Fascingene RNA interference group and non-interference group. Furtheranalysis found that the occurrence and progression of tumor research hasbeen published related genes with FGF-2,PDCD4and ATF3chips appearin our differences in gene chip. [Conclusion]1.FGF-2may be one of downstream signaling molecules promoting theoccurrence and progression of Cholagiocarcinoma.2.PDCD4may be one of downstream signaling molecules promoting theoccurrence and progression of Cholagiocarcinoma.3. ATF3may be one of downstream signaling molecules promoting theoccurrence and progression of Cholagiocarcinoma.
