[Objective] To study the effect of lentivirus-mediated RNA interference(RNAi) of fascin on apoptosis and proliferation of human Cholangoicarcinomacell line QBC939in vitro.[Methods] QBC939cells was conventionally cultured.The experimentalgroup transfected with a lentiviral vector for RNAi targeting the codingregion of human fascin, Nonspecific control group transfected withanother control vector containing a nonsense sequence,and blank controlgroup was treated with the same dose of PBS. The vectors weretransfected into QBC939cells, and fascin expression in the cells wasdetected by RT-PCR and Western blotting. The proliferation andapoptosis of the transfected QBC939cells was evaluated using flowcytometry.[Results] Compared with the control vector, fascin-RNAi-LV resulted inobviously reduced expression of fascin protein, lowered proliferation and inducted apoptosis of QBC939cells (P<0.05).[Conclusion] Lentivirus-mediated nestin RNAi can specifically inhibitFascin expression to cause decreased cell proliferation of humanCholangoicarcinoma cell line QBC939,and can promote apoptosis ofQBC939.
