Objective: Protocadherin-20(PCDH20) belongs to the δ1subgroup ofthe protocadherin subfamily. The human PCDH20gene, also known asPCDH13, is located at13q21.2. Our previously study demonstrated theepigenetic inactivation of PCDH20by promoter methylation and its role asa tumor suppression. In the present study, we further study PCDH20genefunction studies in NPC cell lines, as well as to study its mechanisms.Methods: using Colony formation assay to detect proliferation. Celltransfection techniques, cell cycle analysis was detected by flow cytometry,Scratch wound assay and matrigel invasion assays to test cell migration andinvasion. Applications Western Blot and immunofluorescence detect theexpression levels of PCDH20and relative mechanisms proteins.Results: Over expression of PCDH20inhibits clonogenicity of CNE-1cells and induces G1cell cycle arrest. Over expression of PCDH20decreased the expression of cyclin D1, and up regulate p21protein. CNE-1cell proliferation, motility and invasiveness significant inhibit afterPCDH20over expression. PCDH20antagonized Wnt/β-catenin signalingpathway, thereby inhibiting cell proliferation from the genetic level. Conclusion: The study found that, the percentage of cells in the G1phase was increased in PCDH20-overexpressing CNE-1cells comparedwith control vector cells and CNE-1cells, and proliferation slowed, and therelative cell cycle protein obviously changed. This suggests that PCDH20inhibits CNE-1cells proliferation, further antagonized Wnt/β-cateninsignaling pathway, leading to pathways downstream genes: p-GSK-3β、CCND1、 c-Myc protein expression of CD44upregulated, β-catenintransferred from the cell membrane to the nucleus. While cell migrationand invasion weakened. Thus plays an important role in the development ofNPC.
