Effects Of Langchuangjing Granule On Treating The Prethrombotic State Of MRL/lpr Mice Aggravated By Glucocorticoids

Objective:To observe the curative effect and to explore the possible ways of Langchuang-jing Granule(LCJG) on treating the prethrombotic state of MRL/lpr mice caused by the treatment of glucocorticoids.Methods:36MRL/lpr mice,which were8weeks old females,were divided into model group,control group and experimental group randomly,fed with mid fat diet.12mice in model group were gavaged by distilled water at doses of0.1ml/10g,12mice in control group were gavaged by prednisone suspensionat at doses of5mg/Kg,and12mice in experimental group were gavaged by prednisone suspension(5mg/Kg) and LCJG at doses of5.9g/Kg.These three groups were fed with6weeks.Observed the general conditions and symptoms of those mice.At the end of the experiment, tested the serum leptin level, C-Reactive protein level,anti-ds-DNA level and24hours urinary protein of those mice.Calculated the thymus and spleen indexs of those mice.Observed the pathomorphology of the descending aortas and kidneys by HE stain.And observe the deposition of matrix metalloproteinase-9of the descending aortas by immunohistochemistry,also for the immune complex of the kidneys.Results:The symptom expressions of those mice with no differences.The thymus and spleen indexs of the experimental group were both decreased when compared with the other two(P<0.05).The serum leptin level,C-Reactive protein level of control group rised,while experimental group decreased when compared with model group (P<0.05).The24hours urinary protein and serum anti-ds-DNA level of experimental group and control group were both decreased,but the experimental group were more distinctbal,the differences were statistically significant (P<0.05).Results from the HE stain and immunohistochemistry of the descending aortas showed no significant difference among the three groups,also of the kidneys HE stain. While,immunohistochemistry tests show that the deposition of complement C3and IgG in both control group and experimental group kidneys were less than the model group,and the experimental group is even more obvious.Conclusion:LCJG can decline the serum leptin and C-Reactive protein level of the MRL/lpr mice.It can also help prednisone reduce the leakage of urine protein,reduce the depositions of immune complex in kidneys,so attenuate renal injury,reduce the disease activity.Additionally,LCJG can regulate the functions of immunological organs. So LCJG can relieve the prethrombotic state which was aggravated by the use of glucocorticoids.In a word,LCJG may can increase the therapeutic effects and reduce the side effects of prednisone.
