The Experimental And Clinical Research Of Effects On Anti Breast Cancer Patients During The Perioperation Period With Traditional Chinese Decoction Of Qihuangjianji

Objectives:1. To study the Observation of Qihuang Decoction on MCF-7breast cancer cells and vascular endothelial cells of neonatal HUVEC and explore the influence mechanism on tumor angiogenesis;2observation of Qihuang Decoction on patients with breast cancer during change, angiogenesis serological operation of clinical efficacy and the side on the angiogenesis inhibitory effect. Methods:1according to the cell culture cultured MCF-7cells, HUVEC cells, were divided into five groups,(1) of vascular endothelial cells in the blank group;(2) breast cancer MCF-7cell culture supernatant of HUVEC cells after the action of oxidative stress;(3) the culture supernatant of MCF-7cells stimulated group;(4) HUVEC cells with MCF-7cells culture supernatants stimulated+Astragalus Decoction group;(5) the oxidative stress of MCF-7cell supernatant+Astragalus decoction. MCF-7oxidative stress culture:using hydrogen peroxide reagent is added to the conventional cell culture dish, to simulate stress state of cell oxidation; take MTT cell proliferation inhibition rate and detection, the optimum concentration required for proliferation of Qihuang Decoction on cell in this study, and according to the experimental results, the cell proliferation inhibition rate of Qihuang decoction is in a concentration dependent manner; in accordance with the PCR detection, the drug treated cells, the expression changes of HIF-a mRNA detection, at the same time using the Western blot method of HIF-α, NF-κB protein expression changes;2clinical study:collected in30cases of breast breast cancer patients in our hospital,30patients were divided into two groups,15cases in the control group,15cases in experimental group, the patients in the experiment group,1days before the operation until14days after operation, taking Qihuang decoction, two times a day,100ml each time, while the control group didn’t take Qihuang decoction, in the day before operation after operation the fourteenth day to the two groups of patients with blood examination, preoperative blood is scheduled for the first day, fourth day after surgery, after seventh days and fourteenth days after surgery.Conclusions:1of Qihuang decoction can inhibit MCF-7and HUVEC cell proliferation and inhibits hypoxia inducible factor HIF-α and NF-kb thereby inhibiting angiogenesis;2of Qihuang decoction can improve the clinical symptoms and the peri operation period of serological indexes of patients with breast cancer.
