Objective:Comparison of the physical and chemical properties that prepared by freeze-drying and calcination,Preliminary evaluation biocompatibility of Freeze-dried bone.Methods:Using fresh sheep spine trabecular bone cut into5×5mm, removal of soft tissue and put into methanol/chloroform (3:1),let the Sheep cancellous bone t-reated with chemical method was placed into the muffle furnace,rise1000℃slow and calcined2hours to made calcined bone.Another group will be chemically trea-ted sheep cancellous bone into frozen at-80℃4weeks.Then dried in a vacuum instr-ument prepared freeze-dried bone,60Co irradiation sterilization,let the sheep cancellous bone that throw ultrapure water rinse as a control group.After histological,determi-nation of porosity,EDS elemental analysis,diffraction analysis and mechanical testing, Preliminary analysis of biological performance lyophilized sheep cancellous bone s-caffold evaluation.After MTT,Heat experiments,Acute toxicity test,Intradermal irritati on test, Preliminary analysis of biological performance evaluation lyophilized sheep cancellous bone scaffold. Results:Scaffolds were retained porous three-dimensiona1structure similar with human bone tissue,by microscopic observation,their framewo rk intact,have smaller pores(55-650um),and higher porosity(65%-80%), the scaffold after calcining toughness decrease and brittleness significantly increase.After freeze-drying proces mechanical properties decreased obvious.after diffraction analyze their main components are hydroxyapatite.But calcination also contain small amounts B-TCP,EDS analysis is confirmed the calcium and phosphorus of the three groups were close to the body’s ratio.By biocompatibility testing,lyophilized group scaffold no cytotoxicity,no acute toxicity and heat reaction,intradermal irritation test normal, Chemical group have cytotoxic and mild acute toxicity,are caused by heat and mild irritation.Conclusion:The physical and chemical properties of scaffolds that prepared by freeze-drying and calcination achieve the requirements of bone tissue engineerin-g scaffolds, the freeze-dried Scaffolds have good biocompatibility,Reached the biolo-gical scaffold safety standards...
