ObjectiveTo detect the expression of Survivin and β-catenin in human colorectaladenocarcinoma,colorectal adenoma and normal colorectal tissue(more thanfive cetimetre far away from colorectal adenocarcinoma tissue).Morever,toexplore the correlation between the expression of Survivin and β-catenin andthe clinical pathological parameters in colorectal adenocarcinoma.Analyze therelationship of Survivin and β-catenin in colorectal adenocarcinoma,which isoffering the theory to the diagnosis and treatment for colorectaladenocarcinoma.MethodsUsing the immunohistochemistry PV-9000to detect the expression ofSurvivin protein and β-catenin protein in72cases of colorectaladenocarcinoma,30cases of colorectal adenomas and30cases of normalcolorectal tissues.Western blotting was used to analyze the level of Survivin andβ-catenin protein expression in30cases colorectal adenocarcinoma andadjacent mucosa tissues.Results1Immunohistochemistry results reveal that:①The positive rate of Survivinexpression in colorectal adenocarcinoma is76.4%(55/72),in colorectaladenomas is30.0%(9/30),in normal adjacent mucosa tissues is10.0%(3/30).Obviously,the positive rate of Survivin in colorectaladenocarcinoma was higher than in colorectal adenomas and normal colon tissues,and the difference was statistically significance(P<0.05).②The positiverate of Survivin in colorectal adenocarcinoma was correlated with the depth ofinvasion,differentiation,Dukes stage and lymph node metastasis.The positiverate of Survivin in serosa invaded is higher than non-serosa invaded,there is asignificant difference(P<0.05).The positive rate of Survivin in low-differentiationis clearly higher than high-differentiation and mid-differentiation, and thedifference was statistically significance(P<0.05).The expression of Survivin inDukes A/B stage is lower than C/D stage,there is a significant difference(P<0.05).Futhermore,the expression of Survivin in colorectal adenocarcinoma withlymph node metastasis is higher than no lymph node metastasis, and thedifference is statistically significance(P<0.05).③The ectopic expression rate ofβ-catenin in colorectal adenocarcinoma is higher than colorectal adenomas,thedifference is statistically significance(P<0.05).④The ectopic expression ofβ-catenin is correlation with the depth of invasion,differentiation,lymph nodemetastasis and Dukes stage(P<0.05).The ectopic expression of β-catenin inserosa invaded is higher than non-serosa invaded;The ectopic expression ofβ-catenin in low-differentiation is higher than high-differentiation andmid-differentiation;The ectopic expression of β-catenin in Dukes A/B stage islower than C/D stage; The ectopic expression of β-catenin in colorectaladenocarcinoma with lymph node metastasis is higher than no lymph nodemetastasis, and the difference is statistically significance(P<0.05)⑤There ispositive correlation between the expression of Survivin and ectopic expressionof β-catenin in colorectal adenocarcinoma(r=0.568,P<0.05).2Western blot:The protein quantity of Survivin and β-catenin are higherthan normal adjacent mucosa tissues and colorectal adenomas,the differenceis statistically significance(P<0.05).Conclusion1It presents that Survivin may be play an important role in the developmentof colorectal adenocarcinoma. 2It prompts that β-catenin may be accelerate the development of colorectaladenocarcinoma.3There is positive correlation between the expression of Survivin andectopic expression of β-catenin in colorectal adenocarcinoma,they promote thedevelopment of colorectal adenocarcinoma,contribute to the biological behaviorof neoplasms for deeper research.
