Objective:To acknowledge the association of polymorphisms of klotho gene with low bone mineral density in elderly Uyghur and Han people, and explore the distribution differences of polymorphisms of the gene in ethnic people. Methods:The study adopts a case-control method. A total of324elderly Uyghur and Han patients (≥60years old) were selected from cadre wards of the First Affiliated Hospital of Xinjiang Medical University between October2011and August2013.Lexos dual energy X-ray absorptiometry (DEXA, DMS Co. Ltd, France) was used to check BMD (bone mineral density) of lumbar spine and hip. All the patients were divided into the low BMD case group and control one. Genotype of klotho G395A and F352V polymorphisms were done by multiplex SNaPshot reaction. Results:(1)Both of SNP exist polymorphisms. The distributions of gene polymorphisms in Uyghur and Han people accord with Hardy-Weinberg equilibrium.(2) In case and control groups, the distributions of klotho gene polymorphisms are no statistically significant difference (P>0.05). Overall comparison between the Uygur and Han people, the distribution of G395A polymorphisms is no significant difference (P>0.05); While the TT genotype and T allele frequencies are lower in Uygur national minority than Han crowd (P<0.05), and the TG and GG genotype and G allele frequencies are higher in Han (P<0.05). The diffusions of klotho gene polymorphisms in male and female are no difference (P>0.05).(3) Logistic regression analysis displays that the klotho gene polymorphisms are not associated with low BMD, and female and hypertension are the independent risk factors that affects BMD. Conclusion:G395A and F352V polymorphisms of klotho gene are not associated with low BMD; and the distribution of F352V polymorphisms in Uyghur and Han people exists differences.
