Objective:To observe the effects and mechanism of retinal apoptosis and VEGF、 PEDF factor expression of ozone therapy in the treatment of diabetic retinopathy in rats.Methods:Experimental study.70male SD rats,selected10randomly as normal control group,the rest60feed with high fat and sugar diet, Intraperitoneal injection of streptozotiein(STZ) to induce diabetes.55success, divided randomly into three groups: model control group(n=18, no processing, oxygen intervention group (n=18), give oxygen enema and ozone therapy group (n=19), given ozone enema,1times per3days for4weeks, anesthesia all rats and picked eyes on both sides, TUNEL method to detect the cellular and apoptosis apoptosis index (AI). Detect VEGF expression by Immunohistochemical method. Real-time reverse transcription polymerase chain reaction (RT-PCR) was used to measure the mRNA expression of VEGF and PEDF. Results: TUNEL method express apoptotic cells mainly in the ganglion cell layer, the kernel layer and vascular endothelium. Ozone therapy group amount more than the normal control group, less than the model control group and the oxygen intervention group, apoptosis index differences were statistically significant(F=79.293; P=0.000). The results of Immunohistochemical staining with VEGF express mainly under the internal limiting membrane, RGCs layer and inner plexiform layer, outer plexiform layer and around blood vessels. Negative or weakly positive in normal control group, VEGF expression in ozone treatment group position same with the model control group, but is less than the model control group and intervention group of oxygen. Four groups of retinal RT-PCR test result VEGF mRNA relative expression in the difference was statistically significant (F=15.515; P=0.000). ozone therapy group VEGF mRNA expression group was lower than that of model control group and oxygen intervention group, but still higher than in N group, the difference was statistically significant (P=0.000). Four groups of retinal PEDF mRNA no statistically significant differences in the expression of quantity results (F=2.773; P=0.058). Conclusion:Ozone treatment can reduce early diabetic retinal cell apoptosis, and the secretion of VEGF, may have protective effect of PEDF, at the same time the development of early diabetic retinopathy progression may have the effect of the delay.
