SanZi YangQin decoction, recorded in a book of Ming Dynasty named HanShiYiTong,is used to treat chronic cough and chronic bronchitis, especially for bronchial asthma. Inthis paper, a new cataplasm was developed to substitute the traditional decoction, and its invitro transdermal behavior was investigated. The research work includes:i) To optimize the extraction process of Sanzi. With contents of sinapine thiocyanateand rosmarinic acid, yield of extraction as indexes, orthogonal test method was used tooptimize extraction procedure. Finally, the optimal procedure was verified by contents ofsinapine thiocyanate and rosmarinic acid, and asthma experiment induced by histamine onguinea pigs. The optimal extraction procedure was as follows: the medicinal materials wereextracted by70%ethanol which was equal to10fold of the amount of the medicinalmaterials for three times,1h each time.ii) Optimization of Sanzi cataplasm. Firstly, single factor experiment design was usedto determine the amount of excipients. Subcequently, central composite design was used tooptimize the foemulation of blank cataplasm, with initial bonding strength, endurancebonding strength and comprehensive sense as indexes. Then, optimal Sanzi cataplasm wasobtained by slightly adjusting the amount of excipients to overcome the decrease ofviscosity and oil penetration, which coursed by the addition of Sanzi extract into blankcataplasm.The optimal formulation of Sanzi cataplasm was as follows:1.85g of NP-700,0.6g of K-90,0.13g of dihydroxy aluminum,2g of kaolin,0.08g of tartaric acid,0.02gof EDTA,10g of glycerine,3.5g of Sanzi alcohol extract. iii) Transdermal permeation of Sanzi cataplasm and optimization of penetrationenhancers. Semi-permeable membrane method was used to determine the invitro release ofSanzi cataplasm. The effect of different penetration enhancers on percutaneouse permeationof sinapine thiocyanate and rosmarinic acid through skin was investigated.The resultsshowed that drug release from Sanzi cataplasm was accordance with zero order releasekinetics.The release rate of sinapine thiocyanate and rosmarinic acid were constant with12h, which were37.91μg·cm-2·h-1, and14.30μg·cm-2·h-1, respectively. The penetrationeffect of different penetration enhancers on sinapine thiocyanate and rosmarinic acid was asfollows:2%Azone>1%Azone>3%Azone>1%oleic acid>4%Azone>1%propylene glycol> No penetration enhancers.The results indicated that Sanzi cataplasmwith2%Azone had the highest penetration rate....
