Objectives Use the Nick end labeling method using deoxynucleotidyl transferasemediated (TUNEL) to detect the chorionic villi apoptosis index in drug abortion groupand artificial abortion group. Use the immunohistochemical methods to detect theexpression of bax. bcl-2and survivin in the chorionic villi in drug abortion group andartificial abortion group. Preliminary to study effects of mifepristone on apoptosis andregulating factor in early pregnancy chorionic villi.Methods From2013July to2013November in second hospitals of Qinhuangdao Cityobstetrics outpatient abortion in early pregnancy women, aged20to40years of age,menopause within49days, B ultrasound examination confirmed the diagnosis ofintrauterine pregnancy, no prostaglandin using taboo, and they all signed the informedconsent, the patients according to the intention to divided to drug abortion group andartificial abortion group. Drug abortion group medication method is: the first day and thesecond day morning7points respectively, oral mifepristone75mg, third days morning7points fasting to hospital oral misoprostol600micrograms,6hours observation,According to the gestational sac is discharged into: complete drug abortion group andincomplete drug abortion group each30cases; complete drug abortion group collectionof villi, incomplete abortion group underwent artificial abortion, collect chorionic villi.The control group for artificial abortion group:30cases without preoperative medication,directly artificial abortion, collect chorionic villi.use the Nick end labeling method usingdeoxynucleotidyl transferase mediated (TUNEL) to detect the chorionic villi apoptosisindex in drug abortion group and artificial abortion group. Use the immunohistochemicalmethods to detect the expression of bax. bcl-2and survivin in the chorionic villi in drugabortion group and artificial abortion group; All the experimental results conducted usingSPSS16.0statistical analysis. Measurement data using analysis of variance, rank sumtest with hierarchical data, statistically, that there were significant differences in P<0.05.Results1.The apoptosis of chorionic villi cells occurred mainly in thesyncytiotrophoblast. Apoptosis index in complete drug abortion group was significantlyhigher than that of incomplete abortion group, artificial abortion group were observedunder light microscope (P<0.05); incomplete abortion group and artificial abortion group has no significant difference in apoptosis index (P>0.05).2The expression of Bcl-2ismainly in the syncytiotrophoblast cytoplasmic, cytotrophoblast and interstitial cells wasnot detected. The expression of Bcl-2in complete drug abortion group was significantlylower than that in incomplete abortion group, artificial abortion group (P<0.05); Theexpression of Bcl-2in incomplete abortion group and artificial abortion group has nosignificant difference (P>0.05).3The expression of Bax protein is mainly in thesyncytiotrophoblast cytoplasm. The expression of Bax in complete drug abortion groupand incomplete drug abortion group was higher than that in artificial abortion group(P<0.05); complete drug abortion group and incomplete drug abortion group has nosignificant difference of Bax expression (P>0.05).4Survivin expressed mainly in the cellnuclei of syncytiotrophoblast cells, trophoblast cell nuclei with a small amount ofexpression. The expression of survivin in complete drug abortion was significantly lowerthan that in incomplete drug abortion group and artificial abortion group (P<0.05);incomplete abortion group and artificial abortion group survivin expression has nosignificant difference (P>0.05).Conclusions148hours after the effect of mifepristone, villi apoptosis index increasedsignificantly, that mifepristone may induce apoptosis of villous cells in early pregnancy,achieve the anti early pregnancy objective.2. Bax, Bcl-2were expressed in normal villicells, After48hours of mifepristone treatment, up regulate the expression of Bax in villicells, expression of Bcl-2in villi cells down regulated, mifepristone may through the Baxprotein content increased villus cells, downregulation of Bcl-2protein content in earlypregnant villi cells, leading to balance imbalance Bax/Bcl-2, to achieve the purpose ofanti early pregnancy.3Mifepristone may inhibit survivin gene expression, proliferationand differentiation of the villi further hindered, initiate apoptosis, achieve the purpose ofanti early pregnancy.
