ObjectiveTo investigate partial fucosyltransferase genes expression in myeloid malignancies.The genes include the fucosyltransferase genes in zebrafish/danio rerio (DF1、DF2、DF7、DF8、DF9、DF10、DF11) and the fucosyltransferase genes which constitute A,B,Hantigen or Lewis antigen in human (FUT1、FUT2、FUT3、FUT6、FUT7).MethodsWe used real-time fluorscence quantitative reverse transcription-polymerase chainreaction(FQ RT-PCR)analyzed the fucosyltransferase genes expressions in MYCNtransgenic zebrafish which possesses the characteristics of acute myeloid leukemia andthe FUT1/2/3/6/7expressions in HL-60cells and peripheral blood samples from31acute myeloid leukemia (AML) or myelodysplastic syndrome(MDS) patients and25healthy controls.At the same time, we set wide type(WT) zebrafish as the normalcontrol of MYCN transgenic zebrafish, set the normal CD34(+) cells as the normalcontrol of HL-60cells and set the25healthy controls as the normal control of theAML/MDS patient group.ResultsThe gene expressions of DF7/8/9/10/11in the MYCN transgenic zebrafish andwide type zebrafish are4.858±0.203vs1.005±0.010、0.143±0.022vs1.040±0.184、0.363±0.008vs0.970±0.082、0.103±0.013vs0.994±0.042、0.349±0.023vs0.996±0.035(p<0.01). Compared to wide type zebrafish, the significance of DF1/2geneexpression in the MYCN transgenic zebrafish was not significant(p>0.05).The gene expressions of FUT1/2/3/6/7in the HL-60cells and its normal controlgroup are0.30±0.03vs1.08±0.06、0.68±0.05vs1.20±0.14、0.68±0.04vs1.00± 0.05、0.82±0.03vs1.00±0.05、0.59±0.12vs1.02±0.15(p<0.01).The FUT1/3gene expressions in the patient group and its normal control group are0.85±0.67vs1.31±0.5、0.53±0.48vs1.34±0.69(p<0.05). However, there is nostatistical significance between the FUT2/6/7gene expressions between the two groups(p>0.05). And the differences of FUT1/2/3/6/7gene expressions between AMLpatient(n=13) and MDS patient(n=18) are not significant,either (p>0.05).ConclusionThe DF7/8/9/10/11gene expressions in the MYCN transgenic zebrafish showedsignificantly different from the wide type zebrafish. Compared to the control group, theDF8/9/10/11mRNA level was significantly decreased, only with a high mRNAexpression of DF7. The gene expressions of the FUT1/2/3/6/7showed different degreesof weakening in HL-60cells and the patients with AML/MDS,suggesting somepossible correlations between the gene expressions of fucosyltransferase genes andmyeloid malignancies.
