Objective:The present study aimed at investigating the effect,which madeby several dry needles of inner heating with different diameters and stitches,onmuscle regeneration and repair of the chronic skeletal muscle injury inrabbits,by comparing myofascial microvascular blood flow、 muscle tension andother indicators before and after treatment, providing the basis for itstherapeutic mechanism of experiment on the study in the selection of optimumdiameters and stitches of dry needles of inner heating.Methods:1、grouped:42rabbits were randomly divided into experimental group (28),injury control group and normal control group (14). In experimental group andthe injury control group, The hitting installation is used to make dull injury50times respectively on rabbits’ biceps flexor cruris after half a month as a model ofchronic soft tissue. Normal control group was not injured rabbits.2、 Treatment: There were12needles,length was5cm, heated42℃and20minutes after piercing the skeletal muscle with different diameter and stitchlength and each of the horizontal row and the longitudinal row were3needlesand4needles. According to the different combinations of the dry needle of innerheating with two diameter (1.1mm,0.5mm) and stitch length (0.6cm,1.0cm), theexperimental group was divided into:1.1-0.6cm group (n=14, left leg),1.1-1.0cm group (n=14, right leg),0.5-0.6cm group (n=14, left leg),0.5-1.0cmgroup (n=14, right leg). 3、 The blood flow velocity was observed before and after therapy.Thenumber of the muscle tension (Force-Displacement Distance) should bemeasured not only before the treatment of dry needle of inner heating but alsoafter the treatment at2d,7d and14d。 Rabbits were executed,and the muscletissue of rabbits in different groups are observed by naked eyes and HE stain bymicroscope after the treatment at2d,7d and14d. The volume of MyoD andmyogenin were investigated by immunohistochemistry and analyzed at the samepoints of the time.4、 Statistical analysis The measurements of blood flow and muscletension were determined using ANOVA and analysis of variance aboutfactorial design; MyoD and myogenin protein expression values using bargraphs for statistical description.Results:1、 Change of the muscle tension:(1) Change of the muscle tension before the treatment:Muscle tension waslower in the normal group than the model group before treatment,P<0.01.There was no significant difference of the muscle tension between themodel group and the group treatment,P>0.05.(2) Change of the muscle tension over time:Muscle tension had no changesin the normal group and model group over time.Muscle tension was decreasedover time in group treatment,P<0.01,and more significant in the group of1.1-1.0cm.(3) The effects of stitch length and needle diameter for muscle tension①When the needle diameter was fixed: The muscle tension of1.1-1.0cm was lessthan1.1-0.6cm group after treatment on14d (P <0.01);the muscle tension of0.5-0.6cm group is less than0.5-1.0cm group after treatment on14d(P <0.01).②When the stitch length is fixed:the muscle tension of1.1-1.0cm group isless than0.5-1.0cm group after treatment on14d(P <0.01).③The muscle tension of1.1-1.0cm group is less than0.5-1.0cm group after treatment on14d (P<0.01);1.1-1.0cm group muscle tension decreases most significantly over time(P <0.01).2、Change of the blood flow velocities: The blood flow velocities of normalgroup,before and after disposal, were no significantly difference. The situationis same as model group,P>0.05.However,the blood flow velocity wassignificantly quicker after therapy than before therapy in the treatment group,P<0.01,and was quickest in the group of1.1-1.0cm, P<0.05.3、 Expression of protein of MyoD and myogenin:The average opticaldensity of MyoD and myogenin presents some trends in each group of treatmentover time (at2day、7day、14day after treatment): The average optical densityof MyoD and myogenin over time showed a trend of decline after rising first,andwere less than each treatment group in model group at each time point.Conclusion:Dry needle of inner heating therapy can speed up the blood flow velocity ofthe myofascial microvascular and could improve the muscle tension in rabbitwith chronic muscle injury.when the needle diameter was1.1mm and the needledistance was1.0cm in therapy of dry needle of inner heating for chronicskeletal muscle injury,the changes were more significantly.Dry needle of innerheating therapy can elevate the level expression of MyoD and myogenin,andimprove the regeneration and recovery after skeletal muscle chronic injury.
