Research On The Correlation Between The Expression Of Heparanase And Syndecan-1 In Gallbladder Carcinoma Cell Lines

Objective: To Discuss the relevance of Heparanase and Syndecan1, and Heparanase-Syndecan1 axis regulation in gallbladder carcinoma’s invasion ability.Method: 1. Construct the eukaryotic expression vector of human Heparanase genes and to obtain gallbladder cancer cell line GBD-SD with transgenic overexpression of Heparanase. 2.Compare syndecan-1 expression and distribution conditions between the Heparanase overexpressed gallbladder carcinoma cells and normal gallbladder carcinoma cells with Western-blot. 3.Compare syndecan-1 expression and distribution conditions between the Heparanase overexpressed gallbladder carcinoma cells and normal gallbladder carcinoma cells with RT-PCR.Results: 1. Successfully constructed eukaryotic expression vector of human genes Heparanase genes and to obtain gallbladder cancer cell line GBD-SD with transgenic overexpression of Heparanase.2.The syndecan-1 protein expression of Heparanase overexpressed gallbladder carcinoma cell lines GBD-SD was significantly lower than that of normal gallbladder cancer cell lines Heparanase, the difference was statistically significant(P <0.05).3. The syndecan-1 m RNA level of Heparanase overexpressed gallbladder carcinoma cell lines GBD-SD was significantly lower than that of normal gallbladder cancer cell lines Heparanase, the difference was statistically significant(P <0.05).Conclusion:1. There is a negative correlation between the expression of Heparanase and Syndecan1 in gallbladder carcinoma cell lines2. In gallbladder carcinoma, the expression of Heparanase and Syndecan1 showing a trend of negative correlation similar to other tumor. Heparanase/Syndecan1 axis in gallbladder carcinoma development also played an important role in the occurrence and development of basic research, the gallbladder carcinoma has opened up a new field, but the detailed mechanism of its effect on gallbladder cancer still needs further research.
