The Effect Of Cellular Immune Function And The Observation Of Curative On Sijunzi Mixture Combined With DC-CIK Cells Therapy In Malignant Tumors

Objective Observe the cellular immune function and the clinical efficacy of Sijunzi mixture combined with Dendritic Cells-Cytokine induced Killer cells (DC-CIK cells) immunotherapy for common cancer patients,to investigate the clinical significance and the application perspective of Sijunzi decoction and mixture in the malignant tumors with cellular immune therapy.Methods Select 40 malignant tumor patients conform to the western medicine diagnostic criteria and Chinese traditional medicine criteria,according to a random number table, randomly divided into control group (20 cases) and treatment group (20 cases).The control group only therapy with DC-CIK cells,the treatment group therapy with DC-CIK cells combined Sijunzi mixture,Sijunzi mixture take 2 weeks for a period of treatment.Before treatment,2 weeks and 4 weeks treatment to observe the change of the cellular immune function in patients,and to detect the difference of TCM symptom scores,KPS grades,tumor markers and the adverse reactions between the two groups before and after 4 weeks treatment.Results1.Flow cytometry showed that two weeks after treatment the control group and the treatment group in peripheral blood the CD3+,CD4+,NK cells were significantly higher than before treatment(P<0.05).The treatment group of the CD4+/CD8+was significantly higher than before treatment(P<0.05).The difference comparison among the two groups NK cells increased significantly in the treatment group than the control group(P<0.05).After 4 weeks of treatment, except the CD3+,CD4+cells of treatment group, control group of CD3+,CD4+ and NK cells and NK cells in the treatment group were lower than in treatment 2 weeks,but compared with before treatment,the control group of the CD3+and NK cells and the treatment group of the CD3+,CD4+,CD4+/CD8+ and NK cells were still significantly higher(P<0.05).Compared with two weeks the treatment group of CD3+cells were increased in the four weeks,the difference comparison among the two groups CD3+ cells increased significantly in the treatment group than the control group(P<0.05).2.6 patients of the tumor markers were elevated in the two groups.The control group had 3 cases in all,2 cases after treatment the tumor markers were declined(one case dropped to the normal range),one case was increased.The treatment group had 3 cases,2 cases of tumor markers were dropped to the normal range after treatment,one case was increased.The remaining patients before and after treatment the tumor markers were all in the normal range.3.4 weeks after treatment the primary symptom scores of eat less,body tired weakness,abdominal distension and the minor symptom scores of abdominal pain,bowel sound in the treatment group were decreased significance than the control group(p<0.05). After treatment the two groups of the total TCM symptom scores were decreased significance(P<0.05),the treatment group decreased more significantly than the control group (P<0.05).The treatment group of TCM syndrome evaluate the effective rate was 94.4%,significantly higher than the control group(P<0.05),the control group effective rate was 75%.4.4 weeks after treatment the both groups of KPS scores were significantly higher than before treatment(P<0.05).The treatment group KPS scores increased more significantly than the control group after treatment(P<0.05).5. There were 5 patients had the adverse reaction of fever in the cell infusion process.The control group included three cases, one case of the body temperature was 38.7℃,one case was 38.8℃,one case was 37.6℃.The treatment group had 2 cases,one case was 37.2℃,one case was 38.0℃.There was no bone marrow suppression,liver and kidney function damage,myocardial enzyme damage occurred.The two groups of the safety evaluation has no statistical significance(P<0.05).Conclusion1.Sijunzi mixture can increase the NK cells and CD3+cells in malignant tumor patients treatment by the autologous DC-CIK cells immunotherapy, further improve the cellular immune function.2.Sijunzi mixture can significantly improve the KPS scores and the traditional Chinese medicine syndrome in tumor patients who have the spleen deficiency syndrome,thus improve the patient’s quality of life.3.Sijunzi mixture combined with autologous DC-CIK cells immunotherapy is safety and effective.
