| Functional constipation is a common and frequently-occurring disease, but with the development of the society,several factors such as people’s life style,diet,mental stress has changed alot, the incidence of constipation increased gradually, It seriously affect the patient’s quality of life. At present,There are many ways to treat constipation.General Surgery of my hospital firstly Use not only the medical treatment method such as biofeedback, fecal transplants, probiotics and soluble dietary fiber but also an innovative method of surgical treatment namedâ€Jinling procedur†in the domestic.Although has certain curative effect,but as a result at home and abroad for the pathogenesis of functional constipation is not fully clear, there are still many problems.Several factors contribute to the movement of colon smooth muscle such as the enteric nervous system,neurotransmitters, Cajal interstitial cells, smooth muscle cells and other cells.At home and abroad research on mechanism of functional constipation is mainly focused on "enteric nervous dysfunction".smooth muscle have certain independent rhythmic, its movement is not completely regulated by the neural factors.Therefore,regulation of intestinal nerve function is not the only treatment of Colon motility disorder in patients with functional constipation.To seek some new therapeutic targets except of the enteric nervous system may provide more safe and effective treatments for functional constipation patients who have gastrointestinal motility disorders.Interstitial cells of Cajal is the pacemaker cells of gastrointestinal tract,Depletion of interstitial cells of Cajal is associated with several gastrointestinal motility disorders diseases such as pseudo obstruction, congenital megacolon and ulcerative colitis.TMEM16 A is found recently expression specificity in interstitial cells of Cajal,and it is a reliable and sensitive marker for detecting changes in ICC networks in humans.It may affect the motility of gastrointestinal tract by interstitial cells of Cajal.Thus,we speculate that the expression of TMEM16 A in patients with intractable chronic functional constipation may be abnormal. TMEM16 A may regulate the colon smooth muscle movement through regulating the ICC.In this study,immunofluorescence staining,RT-PCR and Western blot were performed to detect the expression of protein TMEM16 A in colon. At the same time, we will detect c-kit,which is considered the only reliable antigenic marker for ICC,And then analyze the correlation of TMEM16 A and c-kit in colon.So as to Provide theoretical basis for findingnew targets for diagnosis and treatment of functional constipation patients.Part IExpression of TMEM16 A in the colon of intractable functional constipation patients Objective To investigate the expression of TMEMl6 A in intractable functional constipation patients.Methods(1)A total of 30 patients with intractable chronic functional constipation were selected as experimental group, 30 patients with colon cancer as contr1 ol group.Immunofluorescence staining 〠RT-PCR and Western blot were performed to detect the expression of protein TMEM16 A in colon.(2)A total of 100 patients with intractable chronic functional constipation were selected,and then were divided into three subgroups according to their different age,different duration of symptoms and whether have diabetes.RT-PCR and Western blot were performed to detect the expression of protein TMEM16 A in colon.Results The expression of TMEM16 A in immunofluorescenceã€m RNA and Protein content were significantly lower in the experimental group colon than in the control group colon.The expression of TMEM16 A is less in older patientsã€patients who have a longer duration and patients who have diabetes.Conclusion Both protein content and m RNA expression of TMEM16 A was significantly lower in the intractable functional constipation patients.The expression of TMEM16 A is less in older patientsã€patients who have a longer duration and patients who have diabetes.Expression of c-kit in the colon of intractable functional constipation patients Objective To investigate the expression of c-kit in intractable functional constipation patients and study the correlation of TMEM16 A and c-kit.Methods 1)A total of 30 patients with intractable chronic functional constipation were selected as experimental group,30 patients with colon cancer as contr2 ol group.
