Periprosthetic infection is a very serious complication after total joint replacement.There is no effective method to treat infected artificial joint and its surrounding biofilm,and it brings tremendous pressure to patients, clinicians and social medical institutions.Therefore,the importance of the prevention of infection is self-evident.Because of the fine structure of nanometer level and structured three-dimensional structure, Titanium dioxide nanotubes can be used as drug loading position,meanwhile it has good biocompatibility.Thus the titanium implants carried with antibacterial drugs can be a reliable method for the prevention of periprosthetic adherence of bacteria.It can broaden the thinking and choice for the prevention and treatment of periprosthetic infection.Objective:We load titanium dioxide nanotubes with gentamycin,then in vitro study,we observe the relationship between the loading and releasing behavior of antibiotics and the material microstructure characteristics. We study the bacteriostasis on staphylococcus aureus of titanium dioxide nanotubes loaded with gentamicin in vitro and vivo experiments,then we provide a effective treatment for the prevention and treatment of the periprosthetic infection.Method:1.We use electrochemical anodic oxidation method to make the coating structure of titanium dioxide nanotubes in hydrofluoric acid solution.2.We use the method of negative pressure to load titanium dioxide nanotubes with gentamicin,then we calculate the loading efficiency of gentamicin.The gentamicin release kinetics was investigateds.In vitro experiment,the comparison of the adhesion and growth of staphylococcus aureus between titanium dioxide nanotubes loaded with and withoutloaded with gentamici.Also the titanium dioxide nanotubes loaded with gentamici and without loaded with gentamici was put into the femora of the rat,to see the difference of the inhibition on staphylococcus aureus of titanium dioxide nanotubes loaded with and without gentamici was investigated.Results:1 Titanium dioxide nanotubes can be made by using electrochemical anodic oxidation method.2 Titanium dioxide nanotubes can be effectively loaded with gentamicin,and the release curve of gentamicin can be explosive.3 Titanium dioxide nanotubes loaded with gentamicin have the inhibition on staphylococcus aureus,which is c onfirmed in both vivo and vitro experiments.Conclusion:1 Titanium dioxide nanotubes can be made conveniently and efficiently by using electrochemical anodic oxidation method.2 Titanium dioxide nanotubes can be effectively loaded with gentamicin, and they can provide effective antibacterial efficacy,which can broaden the thinking and choice for the prevention and treatment of periprosthetic infection.
