Prognosis for patients with locally adv anced or metastatic disease is poor(5-year survival ratio<5%).Most patients with pancreatic cancer have no chance by surgery at first visit to a doctor.more low er sensitivity to radiotherapy、and higher chemotherapy resistance.Gemcitabine has been considered standard of care for treatment of advanced pancreatic cancer si nce 1997.gemcitabine was found to improve both median overall survival and cli nical benefit response compared to 5-fluorouracil. With clinical appearance of ge mcitabine resistance,However,Gemcitabine-based regimens failed to demonstrate a significant or clinically relevant improvement in median overall survival over gemcitabine alon e. It is also a pressing need to induce the resistant cell line SW1990/Gemcitabin e for further clarify the resistant mechanisms of pancreatic cancer.ObjectiveTo investigate how to induce the resistant cell line SW1990/Gemcitabine for further clarify the resistant mechanisms of pancreatic cancer,and compare the characteristics between SW1990 and SW1990-GZ.MethodsSW1990-GZ was derived from the human pancreatic cancer cell lines SW1990 by exposing the cells to intermittently increasing concentrations of gemcitabine. The IC50 、resistance index、and growth difference of was observed by MTT assay.Cellular intake of SW1990 and SW1990-GZ was detection by HPLC.Results1. The IC50 of SW1990 and SW1990-Gz were respectively(0.07±0.0021)μg/m L 、(87.5 ± 3.24)μg/m L, Resistance index was 1250;Sensibility to gemcitabine in SW1990-GZ were more lower than that in SW990 cell in different concentration s of gemcitabine.2. Growth rate of SW1990-GZ was slower than that of SW1990 from the growth curve.3. Gemcitabine intake in SW1990-GZ was lower than that in SW1990 in different times.Conclusion1. Human resistant pancreatic cancer cell lines SW1990/Gemcitabine was successfully established.Its resistant characteristics were stable and clear.2. SW1990-GZ grows more slowly.3. SW1990-GZ maybe decrease gemcitabine intake by some unclear pathways.
