Objective:Studies the role of estradiol on human bone marrow mesenchymal stem cells(BMSCs) and BMSCs pretreated with estradiol on the mice liver cirrhosis.Methods:(1)The role of estradiol to BMSCs in vitro:the third passages BMSCs were into control group (estradiol untreated) and experiment group (estradiol treated), to measure the role of estradiol on the proliferation activity of BMSC by MTT. Control group and experiment group were made apoptosis by H2O2, normal BMSCs were blank group, to test the effects of estradiol on BMSCs apoptosis by Trypan Blue staining and RT-PCR(test caspase-3 mRNA). (2) The role of BMSCs to liver cirrhosis in vivo:46 normal male C57BL mice were into blank group (n=10), model group (n=12), BMSCs transplantation group (n=12) and estradiol treated BMSCs (BMSCs+E2) transplantation group (n=12). By intraperitoneally injection CCl4 to make liver cirrhosis model. After 12 weeks, estradiol treated and untreated BMSCs respectively transplanted into mice through tail vein, PBS took the place of CC14 for blank group and model group. After 4 weeks transplantation blood used for ALT、AST test, HE-staining and Masson-staining to assess hepatic fibrosis, RT-PCR to analyze collagen type I (collagen I) expression.Results:(1)In vitro experiment:the proliferation activity of experimental group was higher than control group (P=0.000). H2O2 promoted BMSCs apoptosis, experimental group apoptosis was lower than control group (P=0.000). Compare with control group, caspase-3 mRNA of experiment group reduced (P=0.000). (2) In vivo experiment:Successfully made hepatic cirrhosis model; ALT and AST of model group were higher than BMSCs transplantation group and BMSCs+E2 transplantation group, ALT and AST of BMSCs+E2 transplantation group were lower than BMSCs transplantation group (P= 0.000). Compare with model group, fibres of BMSCs transplantation group and BMSCs+E2 transplantation group were reductive. Fibres of BMSCs+E2 transplantation group were lower than BMSCs transplantation group (P<0.05). The collagen I mRNA of model group were higher than BMSCs transplantation group and BMSCs+E2 transplantation group, The collagen I mRNA of BMSCs+E2 transplantation group were lower than BMSCs transplantation group (P<0.05).Conclusion:Estradiol can improve BMSCs proliferation activity and decrease the number of apoptosis made by H2O2; BMSCs can improve liver cirrhosis, and this effect is stronger use BMSCs pretreated with estradiol.
