ObjectiveFatigue is a condition of subjectively discomfort. Chronic fatigue syndrome (CFS) appears due to lack of timely elimination of fatigue, which is a serious risk to human health. There are limited anti-fatigue drugs in current clinical medicine. Extracts from natural products with fewer side effects and without any harmful stimulant ingredients have become a research hotspot in recent years.Millettia Speciosa Champ. Leguminosae, which has tonic effect and can invigorate bone and muscle, is dried root of leguminous plant of Millettia Speciosa and is widely used to cure weakness after diseases and muscle strain. Moreover, it is also with a highly frequent usage of prescription medicine to reenforce strength. This study was firstly to confirm the anti-fatigue activity of water eatract of Millettia Speciosa Champ. Leguminosae. Two different separative parts of water extract were screened to evaluate which part has anti-fatigue effect. And the effective part of its anti-fatigue mechanism was also studied. This study provided pharmacological basic for the development of antifatigue health food.Methods1. Anti-fatigue effect of aqueous extract of Millettia Speciosa Champ. (Leguminosae)The exhaustive swimming and climbing pole tests were applied to explore the anti-fatigue effect of aqueous extract of Millettia Speciosa Champ. (Leguminosae). (MSE). The mechanism as far as energy metabolism system was also discussed.2. The screening test of anti-fatigue and antioxidant activity in vitro of different parts of MSEThe supernatant (MSS) and precipitate parts were obtained after ethanol precipitation treatment of MSE. The crude polysaccharide part (MSP) was obtained after protein-eliminating treatment of the precipitation part. The exhaustive swimming test was used to screen its effectively anti-fatigue part of these two different parts. Combined with crystal violet and screenning DPPH tests, the antioxidant activity of two parts was compared with each other.3. The mechanism of anti-fatigue effect of MSPBy measuring the blood and muscle biochemical indexs in mice after swimming, the mechanisms of anti-fatigue effect association with the antioxidant effect of MSP was explored.Results1. Anti-fatigue effect of MSE①The results showed that, compared with the control group, the MSE group significantly prolonged exhaustive swimming and climbing time of the mice, suggesting that MSE has anti-fatigue effect.②In order to explore anti-fatigue mechanism of MSE, the biochemical indicators of triglyceride (TG), blood urea nitrogen (BUN), glycogen and lactate dehydrogenase (LDH), creatine kinase (CK) after swimming on mice were measured. The results showed that, compared with the control group, MSE could significantly reduce TG and BUN and increase muscle glycogen content. Compared with the control group, MSE could significantly decrease CK activity, but with no statistical significance of LDH.2. The screening tests of antifatigue and antioxidant activity in vitro of different parts of MSEIn exhausted swimming test, compared with the control group, MSP group could significantly extend the swimming time, but had no significantly prolonged effect of MSS, suggested that the anti-fatigue active ingredients were concentrated in MSP part.Combined with crystal violet and screening DPPH tests, it was found that MSP can play free radical scavenging effect, while MSS has less free radical scavenging effect under the condition of the same concentration. MSP was conformed to have antioxidant effect, suggesting that the antioxidant effect may be part of the important mechanisms in anti-fatigue effect.3. The mechanism of antifatigue effect of MSP①Effect on energy supply:MSP could significantly increase blood glucose as well as muscle glycogen content and reduce urea nitrogen in mice.of metabolites:Compared with the model group, MSP was able to reduce the lactic acid in case to accumulate in the muscle tissue, so as to achieve the purpose of anti-fatigue.③Effect on oxidative stress:The experiments showed that MSP could significantly reduce LDH and CK levels in mice which were the indexs of cell damage, increase superoxide dismutase(SOD) and glutathione (GSH) vitality in mice and reduce malonaldehyde(MDA) content.ConclusionTaken together, this paper confirms the crude polysaccharide part of Millettia Speciosa Champ. (Leguminosae) has obvious anti-fatigue effect. The main mechanism was contributed on the regulation of energy metabolism and antioxidant function:To increase energy reservation and blood sugar for energy supplement, reduce metabolites which does damage to the body, strengthen the antioxidative effect to scavenge free radicals produced during exercise.
