| Aims It is to observe the influence of res on memory in Mice with normal mice.Memory acquired disorder mice and Alzheimers disease( AD)mice.Preliminary evaluation of the development application prospect of Resveratrol in prevention and treatment of AD. Methods 1.Effect of resveratrol on the function of learning and memory in healthy miceRandomly assign 50 kunming mice into 5 groups,and they are normal control group,brain convalescent positive control group,trsveratrol in group with high concentration,medium concentration and low concentration,and lavage 8 days contantly.Do the maze experiment in Y type maze 30 min after dosing on the 8t h day.。 In the 30 times reaction jump of one experiment day,put the correct jumping frequency as a wrong result;do test repeatedly after 24 hours,and record the number of wrong jump asmemory performance.Randomly select a mouse from each group in each batch test,dose them 30 min before training,and operate in parallel.Put the first selection of 6 mice inside the grid of the diving platform instrument during the test,let them adapt to the environment in 3 min and set up an electric circuit,mice would jump on the platform in order to avoid electric shock when they felt the energization.Mice feet come into contact with the cooper gate and get an electric shock that being regardrd as wrong reaction.Train 5 min and record the number of getting an electric shock in 5 min,and do test repeatedly after 24 hours,record the number of getting an electric shock identically,and take it as the test results. 2 Effect of scopolamine on the function of learning and memory of memory disorder model mice60 Kunming mice were divided randomly into 6 groups.andthey are normal control group,brain convalescent positive control group,trsveratrol in group with high concentration,medium concentration and low concentration,and lavage 8 days contantly. Mice in ormal control group were intraperitoneally inject NS,and other groups were given intraperitoneal injection of scopolamine 5mg/kg 20 min after dosing on the 8t h day,and do the Y type maze experiment and the diving platform tset 30 min later,record the learning results.Do test repeatedly 24 hours later,and regard the records as the memory results. 3 Effects of resveratrol on the function of learning and memory of the AD modelmice60 Kunming mice with half malesandhalffemales were divided randomly into 6 groups. and they are normal control group,brain convalescent positive control group,trsveratrol in group with high concentration,medium concentration and low concentration( 7.5mg/kg、15mg/kg、30mg/kg). Laboratory adaptive feed three days later, lavage respectivey with 5 CMC-NA mixed suspension and corresponding drugs(with 0.5% CMC-NA suspension liquid preparation),after gavage 20 times in a row,inject 4μl Aβ25- 35 to the tricorn of the control group, positive control group, high, medium and low dose group(7.5mg/kg、15mg/kg、30mg/kg) of resveratrol model group,and positive control group,and inject 4μl NS to the normal control group.Do the water maze experiment after establishing AD model. Result 1. Effects of resveratrol on the ability of learning and memory of healthymice in Y-LabyrinthCompared with the blank control group,the wrong rate on the ability of learning and memory of brain rehabilitation group has a significant decline;the error rate of the two donot have statistical significance although there is a downward trend when the concentration of resveratrol is 7.5 mg/kg; the ability of learning and memory of mice have a significant downward trend( *p<0.05)when the concentration of resveratrol is 15 mg/kg、 30 mg/kg. 2. Effects of resveratrol on the ability of learning and memory of healthymice in Step down testResults show that, piracetam group and resveratrol in high dose group(30mg/kg) and medium dose group(15mg/kg) can enhance the ability of learning and memory of healthy mice(P<0.05);resveratrol in low dose group(7.5mg/kg) has the trend of enhancing the ability of learning and memory of healthy mice but without statistical significance. 3. Effects of resveratrol on the ability of learning and memory of memory acquired disorder mice in Y-LabyrinthCompared with blank group, effect of scopolamine on the function of learning and memory of memory disorder model mice has significant decline,and medol establishing get success. Piracetam group and resveratrol in high dose group,medium dose group and low group all can decrease the error rate of the ability of learning and memory of mice(P<0.01,P<0.05),and enhance the ability of learning and memory of memory acquired disorder mice. 4. Effects of resveratrol on the ability of learning and memory of memory acquired disorder mice in Step down testResults show that,compared with control group, the error rate of resveratrol on the function of learning and memory of the AD model mice has significant decline; piracetam group and resveratrol in high dose group,中 dose group and low group all can enhance the ability of learning and memory of damaged mice(p<0.05). 5. Effects of resveratrol on the ability of learning and memory of AD medol mice in Step down testResults show that,compared with control group,the plateform time of medol group has significant increase; piracetam group and resveratrol in high dose group(30mg/kg),medium dose group(15mg/kg) and low group(7.5mg/kg) all can decrease the plateform time of mice. 6. Effects of resveratrol on the ability of learning and memory of AD medol mice in spatial probe testCompared with blank group,times of medol group mice spanning the original plateform in 60 s has significant decline;times of mice in piracetam group and resveratrol in high dose group(30mg/kg),medium dose group(15mg/kg) and low group(7.5mg/kg) spanning the original plateform in 60 s has significant increase. ConclusionRes has determinate protective effect on cognitive function in AD mice and can improve health, scopolamine to memory disorder model and AD model of learning and mememory ability in miceImprovement of learning and memory ability in health.Scopolamine to memory disorder model mice and AD mice. |
