Traditional Chinese medicine formula shengmai san(shengmai),has been clinically used in the treatment of acute myocardial infarction,cardiac shock and septic shock and other diseases,studies have shown that shengmai formula can improve the impairment of cognitive function induced by hydrobromic acid scopolamine in mice,but so far shengmai formula has not been developed as a health product for improving learning and memory.Our results will provide theoretical base and reference data for further utilizing shengmai ethanol extracts(SM-EE)as an improving learning and memory health food.In present work,normal Kunming mice were chosen to systematically evaluate on the activity of 9:9:6 SM-EE of promoting learning and memory,and then the correlation between promoting learning and memory function and antioxidant effect of SM-EE were discussed.Furthermore,orthogonal experimental were used to obtain the recipes of pharmaceutical preparation of SM-EE soft capsule and SM-EE granule.Finally,the marker substances of total saponins,lignanoids and flavonoids in 9:9:6 of SM-EE and 3:2:1 of SM-EE have been tested,respectively,and then the correlation between these marker substances and their promoting learning and memory activity of SM-EE have been discussed.The results showed that 9:9:6 of SM-EE significantly promoted learning and memory on a dose dependent manner in morris water maze experiment in normal mice.9:9:6 SM-EE on a dose of 0.1 g/kg i.g.had the same effect of promoting learning and memory as that of oxiracetam on a dose of 0.1 g/kg i.g.Compared with 3:2:1 SM-EE,9:9:6 SM-EE had a better effect,and compared with 9:9:6 shengmai powder on a dose of 1.0 g/kg,9:9:6 SM-EE on a dose of 0.1 g/kg also exerted a better effect.After treatment with 9:9:6 SM-EE,SOD and GSH-Px activity in hippocampal tissue in mice were analyzed,as well as the content of MDA.The results showed that SOD and GSH-Px activity levels had no significant change,so did the content of MDA.This suggested that the effect of promoting learning and memory of SM-EE was not dependent on the antioxidation.Our pharmaceutical preparation experiment results showed that the optimum recipe for SM-EE soft capsule is as follows: the relative density of SM-EE was 1.05;the ratio of SM-EE to diluent agent of PEG-400 was 1:2;the ratio of diluent agent of PEG-400 to suspension agent of span-85 was 98.5:1.5.For SM-EE granules,the ratio of SM-EE powder to diluent dextrin was 1:3,the percent of ethanol and adhesive starch was 60% and 5%,respectively.The analysis result of marker substances showed that there was no difference in the content of total saponins and total lignanoids between 9:9:6 SM-EE and the same concentration of 3:2:1 SM-EE.However,the total flavones had a significant difference between them.This indicated that flavonoids may have a correlation with improving learning and memory activity of SM-EE.
