The extraction process, antioxidant activity and their polybutylcyanoacrylate nanoparticles preparation process of the triterpenoids in Inonotus obliquus were studied in this paper. The Inonotus obliquus total triterpenoids extract poly-n-butyl cyanoacrylate nanoparticles were ultimately successfully prepared.First of all, the reflux extraction method (ultrasonic assisted extraction) were used to extract the triterpenoids from Inonotus obliquus, and the content was determined by spectrophotometric method. On the basis of single factor experiment, with the total triterpenoids yield as the evaluation index, the extraction process was optimized by the three factors and three levels orthogonal experiment design. The optimum extraction conditions were:extraction solvent of isopropanol, the material ratio of 1:10. extraction temperature 80 centigrade, extraction time 2h, extraction times of one. In the experimental verification of the optimal process, the average recovery was 1.473%, and the relative standard deviation RSD was 0.48%. The stable and reproducible result showed that the process was reasonable and feasible. After a series of purification process, the Inonotus obliquus triterpenoids extracts were pale yellow powder or flake, luster, and the purity was 80.56%.Secondly, the triterpenoidsthe obtained from extraction and purification in front were studied in vitro antioxidant activity, with the classic antioxidant VC as control substance, using the chemical simulation system to study the antioxidant activity in vitro. Results showed that the triterpenoids of Inonotus obliquus had a certain reduction capacity, was a good electron donor, with antioxidant activity; they could effectively scavenge hydroxyl free radical, superoxide anion free radical and DPPH free radical, and the scavenging rate and sample concentration were in a dose dependent manner, with the sample concentration increasing, the ability of scavenging enhancing. By comparing with the classic antioxidant VC, it proved that the total antioxidant activity of Inonotus obliquus was better.Again, the preparation of Inonotus obliquus triterpenoids polybutylcyanoacrylate nanoparticles by interfacial polymerization, based on single factor experiments of prescription and process factors, with the entrapment efficiency as the evaluation index, orthogonal design was used to optimize the preparation process. The optimal preparation process were determined as follows:15 mg of Inonotus obliquus triterpenoids extract,50 microlitre BCA monomer,2mL ether were as oil phase; 100 mg poloxamer 188 and dextran 70, dissolved in lOmL distilled water as water. In the room temperature and magnetic stirring condition, the oil phase was slowly added to water, stirring for some time. The IOT-PBCA-NP colloidal solution was with light blue milk, and it needed to be refrigerated.Finally, the quality evaluation results of IOT-PBCA-NP prepared by the optimal preparation process were as follows. Scanning electron microscope pictures showed that the nanoparticles were round appearance, uniform size, good dispersion, no adhesion. Particle size distribution was narrow, and basically assumed the normal distribution; the average particle size was 113 nm, with the polydispersity index PI value of 0.128, they had good stability. Zeta potential was-16.0mV; the average entrapment efficiency was 77.68%. The vitro release test results showed that the Weibull distribution function fit the highest, the drug release curve fitting equation was lnln[1/(1-Q)]=0.6151 lnt-1.4308, and correlation coefficient r2 was 0.9869, with obviously sustained effect. It was basically stable in the room temperature for 30 days. Quality evaluation results showed that the IOT-PBCA-NP optimal preparation technology selected by orthogonal experiment was stable and reliable. In this paper, the stable IOT-PBCA-NP were synthesized by interfacial polycondensation method, oral or intravenous injection. The nanoparticles with small particle size and uniform distribution, can achieve the purpose of slow release without stimulation.
