| Objective:To detect the expression of PRR11 protein in normal pancreatic tissue, adjacent non-cancerous pancreatic and pancreatic tissues, and to analyze the relationship between the expression of PRR11 protein in pancreatic cancer and pancreatic cancer clinicopathological parameters as well as survival time of patients, to explore the role of PRR11 protein in the carciongeresis and development of pancreatic cancer and the relationship with the prognosis of patients with pancreatic cancer.Method:1. Selected 58 specimens from patients who underwent pancreatic surgery in Hunan Provincial People’s Hospital from January 2010 to July 2013, including 6 cases of normal pancreatic tissue,20 cases of adjacent non-cancerous pancreatic tissues,32 cases of pancreatic cancer. Immunohistochemistry (SP methods) was used to detect the expression of PRR11 protein in all 58 tissue specimens, then chi-square test was used to analyze the relationship between the expression of PRR11 and clinicopathological parameters (age, gender, TNM stage, tumor size, tumor location, lymph node metastasis, differentiation) of pancreatic patients.2.32 cases of pancreatic cancer patients were followed-up postoperativly by telephone investigation and outpatient clinic visits.end-points of follow-up was July 1st,2014 or determined by patients death.Results:1. positive expression of PR11 protein was mainly located in the cytoplasm, the positive cells were brown-yellow granules, the positive rates of PR11 in normal pancreatic tissue, adjacent non-cancerous pancreatic tissues pancreatic cancer were 0, 5% and 78.1%, respectively. Results show the pancreatic cancer group has a significant higher expression of PR11 compared with the adjacent non-cancerous pancreatic tissue group (P<0.05), While no significant difference were observed between the adjacent non-cancerous pancreatic tissue group and the normal pancreatic tissue(P> 0.05).2. The positive expression rate of PRR11 protein in pancreatic cancer was associated with the cell differentiation degree of pancreatic cancer and TNM staging (P <0.05), but with the patient’s age, sex, tumor size, lymph node metastasis, and tumor location of independent (P> 0.05).3.32 cases of pancreatic cancer patients, PRR11 protein expression in patients with an average survival time was 9.5 months, PRR11 protein-negative patients the average survival time of 14 months, PRR11 protein-negative patients survive longer than the positive expression, and the difference statistically significant (p<0.05), by Cox model analysis, the degree of differentiation as an independent prognostic factor.Conclusion:1.PRR11 protein expression in pancreatic cancer patients with an average survival time of patients with less than negative expression, suggesting that the protein detected PRR11 prognosis of pancreatic cancer has clinical reference value.2.PRR11 protein expressed predominantly in the cytoplasm, a little expression in the cell membrane.PRR11 was mainly expressed in the cytoplasm, a little expression in the cell membrane, in pancreatic tissue from benign to malignant transformation process PRR11 protein expression levels were increased gradually, and poorly differentiated group PRR11 protein was higher expression than highly differentiated group. PRR11 protein for diagnosis of pancreatic cancer diseases have a certain reference value. |
