Objects:To discuss the relationship between IL-10 and venous ulcer and the differences in the pathogenesis of Venous ulcer. Compare the expression level of IL-10 in the skin of lower limb before and after operations by the therapeutic operations(the long saphenous vein was ligationed and striped and the ulcerated skin was sutured) ulceration and Provide the theory evidence for the surgical treatment of ulceration. Methods:25 patients with Simple varicose veins and 21 pations with PTS treatmented in our hospital ware included and collection their ulceration skin.All patients underwent angiography of lower extremity deep venous to make clear the pathogeny and rule out the artery ischemia, diabetes, infections and autoimmune diseases. All patients were cured by operations(the long saphenous vein was ligationed and striped and the ulcerated skin was sutured) and none used Antiphlogistic medicine. IL-10 was analyzed by immunohistochemistry among ulceration group、ulceration group by sugery a week later and normal skin group(the incision skin of flush saphenofemoral ligation of the major saphenous vein). It is Positive when cell membrane or cytoplasm appeared brown. Each slice was randomil observed 3 fields in the high power field(X400), each field choice 100 cells and found the positive cell. calculate the average rate of positive cells. Expression of cell data positive mean standard deviation in soil(`x±S) to represent, SPSS software for t test, take the significance level alpha =0.05, P <0.05 was considered as significant difference. Results:Through statistical analysis, the expression of IL-10 in ulcer tissues was significantly higher than that in normal skin tissue before operation(P <0.05), the expression of IL-10 after seventh days in the ulcer tissues is lower than before(P<0.05),there is no significant difference in the expression of IL-10 in the pations with PTS or simple varicose veins. The cytoplasm or cell membrane of the cells in the ulcer tissue was stained deeper brown, and they mainly located in the dermis. There are many inflammatory cells in the ulcer, and the vessels is richer than that in the normal skin tissue. The number of positive cells in the ulcer tissue before operation is obviously higher than that after surgery.Normal skin tissue visible a few scattered distribution in the positive cells distributed coloring lighter.There is a few positive cells in the normal tissue and they were dyed slightly. Conclusions:1.The corelation between high expression of IL-10 and venous ulceration is significant.2.There is no difference in the expression of IL-10 in the patients with simple varicosis and PTS, which shows the pathogenesis of the two diseases are similar.3.The level of IL-10 decline significantly after the surgery, so it could be used as evaluation index of postoperative effect.
