Objective: To investigate the role of TGF-β1/Smads pathway in pathogenesis of psoriasis, expecting to provide a certain degree of reference for therapy of this disease.Method: The expression of TGF-β1,Smad2/3 and Smad7 in 58 cases of psoriasis(psoriasis vulgaris(n=19), pustular psoriasis(n=20), erythrodermic psoriasis( n=19)) and normal skin( n=10) was determined using immunohistochemistry SP assay, and then to compare among the groups.Results: In normal skin tissue, the expression of TGF-β1,Smad2/3 and Smad7 was located in the basal layer of epidermis, but weak or absent expressed in the stratum spinosum of epidermis and the papillary layer of dermis. In psoriatic lesion, the expression of TGF-β1 was located in the papillary layer of dermis, but weak or absent expressed in epidermis. The expression of Smad2/3 and Smad7 was significantly reduced in the basal layer of epidermis, and remain weak or absent expressed in the stratum spinosum of epidermis and the papillary layer of dermis. Quantitative analysis: Compared with the normal groups(0.0091±0.0032),The expression of TGF-β1 in the psoriasis groups(0.0209±0.0018), erythrodermic groups( 0.0182±0.0023) and vulgaris groups( 0.0254±0.0033) was slight up-regulation(P<0.05), but no statistical difference to pustulosa groups(0.0144±0.0035), and no significant difference among the three types of psoriasis was detected. The expression of Smad2/3(psoriasis groups 0.0003±0.00006,erythrodermic groups(0.0004±0.00009),vulgaris groups(0.0002±0.00004), pustulosa groups(0.0004±0.00016), normal groups(0.0017±0.00038) and Smad7( psoriasis groups 0.0002±0.00003, erythrodermic groups(0.0002±0.00007), vulgaris groups(0.0001±0.00003), pustulosa groups(0.0002±0.00007) and normal groups(0.0017±0.00057) was significantly down-regulated in psoriatic lesion, but no significant difference among the three types of psoriasis was detected.Conclusion: ①The abnormal distribution of TGF-β1 may involved in the pathogenesis of psoriasis. ②The damaged of TGF-β1/Smads pathway may play an important role in the proliferate of epidermis in psoriatic. ③The damaged of TGF-β1 /smads pathway may recede the function of TGF-β1 for inhibiting shin inflammatory, thereby promote the develop of inflammatory.
