Objective:Through animal experiment alone or in combination with antitumor drug pemetrexed, lobaplatin and sunitinib acts on human osteosarcoma cell line 143B in nude mice animal model, investigate the inhibiting ability of osteosarcoma in nude mice of the drugs, to find a new safe and effective therapy for clinical chemotherapy of osteosarcoma.Methods:To establish human osteosarcoma in nude mice animal model with, fluorescent protein markers. After the success of the modeling of the nude mice,30 nude mice were randomly divided into 6 groups, were given different medications, saline control group (G1), lobaplatin group (G2), pemetrexed group (G3), lobaplatin and pemetrexed group (G4), sunitinib group (G5) and lobaplatin and sunitinib group (G6).The corresponding drugs were given to the therapy after 2 days of tumor formation of the nude mice. Tumor volume was measured by fluorescence photography every week. Ended the experiment after using the drugs for four weeks. The nude mice were killed to obtain tumors, tumor weight was measured, and the obtained data were statistically analyzed. The tumor specimens were made HE staining to observe the morphology of tumor tissue under the microscope.Results:Through the measurement, from the second week of the experiment, tumor volume of nude mice were significantly lower in the lobaplatin and sunitinib group (G6) than in the control group (Gl) (P< 0.05). From the third week of the experiment, tumor volume of nude mice were also significantly lower in the lobaplatin and pemetrexed group (G4) and sunitinib group (G5) than in the control group (G1) (P< 0.05). At the end of the experiment we measured the weight of tumor, tumor weight of nude mice were significantly lower in the sunitinib group (G5) and lobaplatin and sunitinib group (G6) than in the control group (G1) (P< 0.05). Observation of tumor tissue under the microscope, lobaplatin and pemetrexed group (G4), sunitinib group (G5) and lobaplatin and sunitinib group (G6) compared with the control group (G1), showed apoptosis and necrosis of tumor cells in different extent.Conclusion:In addition to single use of sunitinib, single use of other drugs on the inhibition of human osteosarcoma in nude mice animal model of tumor proliferation is not clear. Single use of sunitinib, combined with pemetrexed and lobaplatin and combined with sunitinib and lobaplatin have evidently inhibitory effects on tumor volume growth in nude mice. Single use of sunitinib and combined with sunitinib and lobaplatin have evidently inhibitory effects on tumor weight growth in nude mice. The experimental results can provide new chemotherapy of osteosarcoma, and can be chosen by patient who has resistance of the first-line osteosarcoma chemotherapy drugs, or can be chosen as a second-line chemotherapy for patients with recurrence and metastasis of osteosarcoma.
