Objective: To study the correlation among plasma homocysteine(Hcy) level and their metabolic enzymes gene polymorphism and CAD in Han Population of Northeast Sichuan,and to investigate whether elevated Hcy、their metabolic enzymes gene MTHFR C677T、MTHFR A1298C、MS A2756 G 、 MTRR A66 G mutation and gene-gene interation 、 geneenvironment interation will increase the risk of CAD.Methods:then Hcy were detected by Homocysteine Assay Kit(Hcy),A newly-developed technology named improved Multiplex Ligation Detection Reaction, i MLDR, was used to detect target genes and SNPs. SPSS13.0 and MDR(Multifactor Dimensionality Reduction) were used to analysis results.Results:1.(1)The plasma Hcy level in the CAD group was15.39±6.89 umol/L,control group was12.90±6.44umol/L,The plasma Hcy level in the CAD group significantly higher than that in the control group, and the difference was statistically significant(P<0.05),OR of Hcy was1.060(95%CI 1.021~1.100) the difference between two groups had statistically significance(P<0.05) 。(2) The plasma Hcy level in stable angina pectoris(SAP) was12.36±4.5umol/L,The plasma Hcy level in ustable angina pectoris(UAP) was14.90±6.29umol/L,The plasma Hcy level in ischemic cardiomyopathy was15.39±6.80umol/L,The plasma Hcy level in acute myocardial infarction(AMI)patients was16.49±7.66umol/L,The plasma Hcy level in AMI patients was the highest, and the difference between AMI and SAP had statistically significance(P<0.05).(3) The plasma Hcy level in 1 vessels lesions was13.87±4.99umol/L,The plasma Hcy level in 2 vessels lesions was15.81±8.14umol/L,The plasma Hcy level in 3 vessels lesions was17.74±7.15umol/L,and the difference between 1 vessels lesions and 3 vessels lesions had statistically significance(P<0.05),indicating that plasma Hcy was related to coronary arteries lesion severity.2.MTHFR C677T、MTHFR A1298C、MS A2756 G and MTRR A66 G respectively had three genotypes in the case group and control group, the difference of genotype frequencies and allele frequency between two groups didn’t have statistically significance(P<0.05),and by MDR we discovered between MTHFR C677T、MTHFR A1298C、MS A2756 G and MTRR A66 G didn’t have interaction and between MTHFR C677T、MTHFR A1298C、MS A2756 G 、MTRR A66 G and smoking、DM、hypertension、 family history、TG、Hcy didn’t have interaction,the difference didn’t have statistically significance(P>0.05)。3.In case group the plasma Hcy level of patients with MTHFR C677 T CC genotype was 13.99±4.77umol/L, the plasma Hcy level of patients with CT genotypes was 15.44±6.25umol/L,the plasma Hcy level of patients with TT genotypes was 19.72±11.51umol/L, the difference between TT genotypes and CT genotype、CC genotypes had statistically significance(P<0.05)。Conclusion:1. Hcy was a risk factors for CAD in Han Population of Northeast Sichuan.2. The plasma Hcy level in AMI patients was the highest in different clinical types of CAD,The plasma Hcy level in 3 vessels lesions was the highest in different vessels lesions,indicating that plasma Hcy was related to myocardial infarction and coronary arteries lesion severity.3.MTHFR C677T、MTHFR A1298C、MS A2756 G and MTRR A66 G respectively had three genotypes in the case group and control group, difference of genotype frequencies and allele frequency between two groups didn’t have statistically significance(P<0.05), between MTHFR C677T、MTHFR A1298C、MS A2756 G and MTRR A66 G didn’t have interaction and between MTHFR C677T、MTHFR A1298C、MS A2756 G 、MTRR A66 G and smoking、DM、hypertension、 family history、TG、Hcy didn’t have interaction.4.In case group the plasma Hcy level of patients with MTHFR C677 T TT genotypes was the highest in three genotypes, the difference between TT genotypes and CT genotype、CC genotypes had statistically significance.(P<0.05)。...
