Objective:thyroid nodule is the clinical common disease, autopsy data showed that 50% of peop le(age 42-91 years old) had thyroid nodules with no clinical symptoms which were greater th an 1 cm[1]. And identification of benign and malignant nodules is an important work of endocrin e doctors, at present we mainly rely on clinical thyroid fine-needle aspiration biopsy (FNAB), ultr asound, thyroid function, signs and family history to distinguish between benign and malignant t hyroid nodules. Thyroid FNAB is particularly important, but for thyroid follicular tumor FNAB a s cytology, unable to extract complete tumor tissue structure, diagnose with difficulty. But with th e development of molecular biology techniques, found that more and more molecular biomarker s may be used inthe identification of benign and malignant tumor of thyroid follicular type. But n ow we have not yet found molecular biomarkers can be used independently for diagnosis. In the se molecular biomarkers:intestinal trefoil factor,(TFF3), chromosome 1 open reading frame, (Clo rf24) to differentiate benign and malignant thyroid nodules have high specificity and sensitivity. T herefore this study use RT-PCR technique in the nucleic acid level to detecte the gene expressio n level of TFF3, Clorf24 in tumor tissues that obtained by thyroid fine needle biopsy of the thy roid,to evaluate whether it can be used clinically in the identification of benign and malignant o f thyroid follicular type. Methods:selection of thyroid nodules tissue specimens(FNAB)from Jinling hospital between Ap ril 2013 and April 2014, a total of 279 cases, including consideration of thyroid papillary carcino ma 1 case, undifferentiated thyroid carcinoma 1 case, fishy follicular type lesionsin 42 cases. A t otal of 44 patients were considered as malignant or suspicious malignant thyroid nodules, suggeste d these patients with surgical treatment. Late follow-up 39 cases of the operation, operative patho logy report tip:27 cases of thyroid follicular adenoma,6 cases of follicular thyroid carcinoma, thy roid papillary carcinoma in 1 case,2 cases of nodular goiter and thyroid anaplastic carcinoma i n 1 case, hashimoto thyroiditis in 2 cases. Stored immediately after extraction in-80℃ refrigerat or. Batches using RT-PCR techniques to extract RNA, and detecte the expression levels of TFF3, Clorf24 and Beta-actin (reference gene) of tissue samples at the nucleic acid leve 1. Expressed as a ACT of relative gene expression. Separately rank test datas ofΔCT for eac h group. (P< 0.05) difference was statistically significant. Analyzed by SPSS 19.0 software. Results:The expression level of TFF3 mRNA was higher in follicular thyroid carcinoma group co mpared to follicular thyroid adenoma group (P<0.05)The expression level of Clorf24 mRNA was higher in follicular thyroid carcinoma group compared to follicular thyroid adenoma group (P<0.05)...
