Tengcha is a wild liane,belonging to Ampelopsis Michx of Vitaceae.With the effect of heat-clearing and detoxifying, it has been used to treat with pyretic fever and pain-swelling of pharynx in the folks. In the southern provinces such as Guangdong, Guangxi, Jiangxi and Fujian, it is distributed widely with rich resources. Ampelopsis is a extracted part of flavonoids from Tengcha in Fujian,with an average content over 15 percents.This paper focus on the apoptosis mechanism of LNCaP cells by Ampelopsis. It has been witnessed by many experiments that these compounds have active function in vitro on many cancer cells.Nowadays, prostate cancer is a very common disease in male urogenital system,meanwhile the second killer of all the fatal tumors. China has witnessed a dramatic growth of prostate cancer, which brings much harmness to the health of the publics.We have conformed by the experiments that Fujian Ampelopsis has proliferation inhibition and apoptosis effects on many prostate cancer cells, but the mechanism is still undefined.This paper verified the proliferation inhibition and apoptosis effects on LNCaP cells of Fujian Ampelopsis and discussed the mechanism of apoptosis.Cell morphology observation and MTT assay were used to study the inhibitory effects of Ampelopsis on LNCaP cells. The results showed that Ampelopsis inhibite the human prostate cancer cells LNCaP distinctly, with dose-dependent manner. The IC50 was 28.45μg/ml.By Annexin V/PI fluorescent staining of flow cytometric method, the result showed that Ampelopsis can induce Lncapcells apoptosis, with dose-dependent manner.Western Bloting was used to observe the expression of the proteins in PI3K/Akt signal pathway and study the apoptosis mechanism of LNCaP cells by Ampelopsis. The results showed that Ampelopsis can control PI3K/Akt signal pathway,with the lower expression of p-Akt and Bcl-2, then induce LNCaP cells apoptosis.The conclusion could be drawn that Ampelopsis has proliferation inhibition and apoptosis inducement with LNCaP cells. And this function maybe concerned with the regulation of PI3K/Akt signal pathway. These experimental results could be offered for the further research and development of Ampelopsis.
