The Experimental Research Of Grape Juice On LNCaP Human Prostate Cancer Cell Growth In Vitro

Objective To study the effect of Grape juice on cell growth and apoptosis,and the expression of survivin and caspase-3 mRNA in human prostate cancer LNCaP cells.Methods Different concentrations of grape juice(32,64,128?l/ml)were added in prostate cancer LNCaP cells for 24 h,48 h,72 h respectively.Cell inhibition effect was measured by MTT assays.Cell apoptosis was analyzed by flow cytometry.Expression of the survivin and caspase-3 were analyzed by real-time RT-PCR.Results Each group of Grape juice has obvious inhibitory effect on prostate cancer LNCaP cells,rendering time dose dependent.With the Flow cytometry instrument test results to determine the cell apoptosis,the apoptosis cells of all concentration Grape juice were significantly higher than that of control group.Real-time RT-PCR determination of mRNA levels in LNCaP cells in prostate cancer,the caspase-3 mRNA levels in each experiment group increased significantly compared with control group(P<0.05),While the survivin mRNA levels were significantly lower compared with control group(P<0.05).Conclusion Grape juice can induce the LNCaP cells apoptosis possibly through decreasing the mRNA expression of survivin,improving the mRNA expression of caspase-3.
