Objective:In this study we investigate the expression of IDOl in bone marrow of AA, to explore the cellular immune condition of AA, and also to analyze the the significance related to the IDO1 and changes of immune function in AA patients, analysis the influence of 1ST on immune function.To explorethe role of IDO1 in the pathogenesis of AA.Methods:Collected during 2012 December to 2014 December 16 cases of newly diagnosed AA and 8 healthy controls,all cases were confirmed by blood,bone marrow and other checks and meet the diagnostic criteria of 《Expert Consensus statement on Aplastic anemia diagnosis and treatment)), We used RT-qPCR to detect the expression of IDO1mRNA with AA patients、after thetreat-ment by 1ST, and 8 normal controls, and compare with the same period lymp-hocyte subset and Tregs cells, to observe change of cellular immunity.Results:(1) There were high expression of IDO1mRNA in AA patients b-one marrow(0.46±0.20), obviously higher than control group (RQ=6.01); (2)The the expression of IDO1mRNA in patients after three months immunosuppressi ve treatment were significantly decreased compared with the pretherapeuticpatie nts (RQ=4.62);(3)Expression of IDO1mRNA had no relationship with the s-ame period lymphocyte subset (CD3+T cell, CD3+CD4+T cell、CD3+CD8+T cell、C D16+CD56+NK cell、CD16+CD56+NK cellin AA patients;Theexpressionof ID-O1 mRNA of aplastic anemia patients were negatively correlated to the proportion of Treg cells, (r=-0.403, P<0.05). (4)The percentage of CD4+cells withn-e wly diagnosed AA patient was less than controls,The percentage of CD8+cells were significantly higher than controls, the difference was significant (tl=5.43, t2 =16.40, P<0.01);CD4+/CD8+ratio were significantly higher than controls, the d-if ferencewas significant (%=7.32, P<0.01);The percentage of CD4+CD25+Treg w ere less than controls, and were significantly increased after treatment,the differenc e was significant.ConcIusion:The expression of IDO1mRNA in AA patients bone arrow was higher than control group, and it was decreased after 1ST. IDO1 detection in AA was helpfulto evaluate immune condition and therapy effect.
