China is the world’s production of persimmon and the country of origin. Persimmon is China’s fifth largest seasonal fruit, yield and planting area are very impressive. Persimmon including lowering blood pressure, anti inebriation, snake venom solution, anticancer and antiviral function. Medical studies and modern food show that nutrients rich persimmon fruit contains tannin, which is the main effect. Tannin is complex and diverse phenolic compounds of a class of a molecule are widespread in the plant kingdom is larger, can be combined with protein, alkaloid. The study confirmed that the tannin is convergence hemostatic, antibacterial, anti diarrhea, protect the mucosa.With the study of tannin structure and chemical composition, more and more domestic and foreign scholars committed to explore its role in clinical diseases.Candidiasis is a common disease in clinical fungal infection. In recent years, due to the application of the abuse of broad-spectrum antibiotics, radiotherapy, chemotherapy and immune inhibitors application, more and more denture, stents, catheters, organ transplantation and so on, lead to oral candidiasis rising incidence. With the use of antifungal agents, resulting in the production of drug-resistant, oral health micro ecological environment has been damaged, thereby increasing the difficulty of treatment of candidiasis. Extraction of effective components from natural plant for the treatment of candidiasis has become the new research trend. Chinese herbal medicine in anti fungi have great advantages, it can regulate the immunity while anti fungal infection, and less adverse reactions, is not easy to produce drug resistance. Because of its powerful tannins are main research object of the scholars at home and abroad, therefore, through the experimental study of persimmon tannin extract on oral Candida albicans inhibition, has positive effect on the treatment of candidiasis.Objective:China is rich in Persimmon Resources, medical research and modern food that contains rich nutrient components in persimmon fruit, which has important health value, tannins have been confirmed as its main efficacy components. Domestic and foreign scholars to study more and more persimmon tannin, tannin content were compared in this study on different varieties of persimmon fruits, provide the basis for the application of persimmon tannin in stomatology, research and exploration of persimmon tannin plays an active role in.With the rising rate of candidiasis, and the generation of resistance and increase the difficulty of treatment, so finding little drug resistance, anti fungal drugs less toxicity has become an inevitable trend. The experimental study in vitro effect of persimmon tannin water inhibitory effect of extract of Candida albicans and the expression of drug resistance gene CDR1. Provide the basis for the application of persimmon tannin in stomatology, research and exploration of persimmon tannin plays an active role in.Methods:1 Persimmon tannin extract : respectively on the persimmon Gai, persimmon Niuxin, persimmon Big Mopan fruit smash all grinding, soaking in ethanol after ultrasonic extraction, adsorption in addition to sugar, rotary evaporation concentrated solution with distilled water and diluted with AB-8 macroporous resin, polysulfone hollow fiber ultrafiltration membrane ultrafiltration, the molecular weight of persimmon tannin solution 10000, persimmon tannin content determination of water extract liquid tannin using Folin-Denis method, the comparison of the differences of tannin content of different varieties of persimmon fruit.2 In vitro antibacterial test of full persimmon tannin extract on Candida albicans: modified liquid two times dilution method, detection of persimmon tannin extract liquid water minimal inhibitory concentration against Candida albicans, then using reverse transcriptase polymerase chain reaction for the detection of drug resistance of Candida albicans CDR1 gene expression level in different drug concentration after treatment.Results:1 Tannin reference solution at 1.6×10-3mg / m L to 7.2×10-3mg / m L was linear within the range, the standard curve regression equation is y=39.583x+0.013, correlation coefficient R2=0.994. The persimmon Gai tannin content is 202±3.6mg per gram, per gram of persimmon Niuxin tannin content is 73±2.4mg, per gram of persimmon Big Mopan tannin content of 52±3.0mg large.2 Persimmon tannin extract liquid water minimum inhibitory concentration against Candida albicans was 0.1159 mg/m L; with the increase of drug concentration gradient, the expression level of resistance associated Candida albicans CDR1 gene expression level decreased, the concentration gradient of the resistance gene m RNA was completely inhibited in 3.71mg/m L.Conclusions:1 This experiment were extracted from persimmon Gai, persimmon Niuxin, persimmon Big Mopan fruit tannins in full by ultrasonic extraction method, extraction and determination of tannin content in liquid by using the Folin-Denis method, the content of tannin in persimmon fruit compared different varieties: persimmon Gai > persimmon Niuxin > persimmon Big Mopan2 Persimmon tannin water extract in vitro on Candida albicans has certain antibacterial activity and have certain inhibition effect on Candida albicans.
