Objectives To analyze the heme oxygenase-1 gene single nucleotide polymorphisms (SNPs) of rs2071746 and rs2071749, then explore the association between rs2071746 and rs2071749 polymorphisms and hepatitis B (CHB), HBV related liver cirrhosis (LC) and hepatocellular carcinoma (HCC) for providing some new insight in the prevention and treatment of liver disease with HBV infection.Methods In total,111 patiens with CHB,90 patients with LC,267 patients with HCC and 249 healthy controls were involved in this study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing methods were used to analyze polymorphysms HO-1. Haplotypes analysis was used the SHEsis online sofware. The logistic regression analysis was used to assess associations by calculating odds ration (OR) and 95% confidence interval (95%CI).Results1. The genotype distributions of rs2071746 and rs2071749 in four groups were all conform to HWE, the sample of our study was representative and comparable.2. There were three genotypes in rs2071746 polymorphism (TT, TA and AA). The A allele and AA genotype were associated with incrsssed risk of CHB and HCC. There were three genotypes in rs2071749 polymorphism (GG, GA and AA). No association was observed between genetic polymophism of rs2071749 and CHB, LC, HCC (p>0.05).3. When stratified by gander, the A allele and AA genotype of rs2071746 were associated with susceptibility to CHB in male.4. When stratified by ethnicity, the significant statistically association was only detected between all LC and genotypes and allele of rs2071746 in Han population.5. When stratified by age, the A allele and AA genotype of rs2071746 were associated with CHB risk in the age<50 years, and with HCC risk in the age^ 50year.6. When stratified by BMI, the A allele and AA genotype of rs2071746 were incressed risk of CHB in the BMI<24; AT genotype of rs2071746 were associated with LC risk in the BMI≥24.7. When stratified by smoking status,the significant statistically association was only detected between CHB and the A allele and AA genotype of rs2071746 in no smoking population.8. When stratified by alcolhol assumption status, no association was found between the two SNPs and susceptibility to CHB, LC and HCC.9. Haplotypes analysis results shown AG haplotypes increase the risk of CHB and HCC.Comclusions1. The A allele and AA genotype of rs2071746 polymorphism maybe the risk factor for CHB、LC and HCC. But there is significant difference between different gander, ethnicity, age, BMI and smoking status.2. The rs2071749 polymorphism may be not associated with the risk of CHB, HBV related LC and HCC.3. AG haplotypes of rs2071746 and rs2071749 may be risk factor of CHB and HBV related HCC. TG haplotypes may be pretect factor of CHB...
