Objective: For a long time, lymphatic vessel is considered to be the passive participants in the process of tumor metastasis, and simply transport corridor for tumor cells to lymph drainage. With the discovery of lymphatic specific molecular markers, and related research progressing, the credibility of lymphatic biology conclusion has improved, further proof that the relationship and the mechanism between the lymphatic system and metastatic spread is very complicated. D2-40 is a kind of protein molecules expressing in lymphatic endothelial cell, it is found by researchers because of it can specially combine with the carcinoembryonic antigen M2 A expressing in fetal testis and testicular germ cell tumor.At present, most researches consider that D2-40 is only expressing in lymphatic endothelial cells, and not in vascular endothelial cells, is the most effective and specific markers of lymphatic,and the specificity is up to 98%.Micro lymphatic vessel density(LMVD),is the indicators of quantity of D2-40 positive expression,is also a evaluation index of tumour lymphangiogenesis.Observing LMVD can estimate for generation of lymphangiogenesis objectively,and help for assessing the risk of cancer lymph node metastasis and prognosis. RUNX3 gene is a new tumor suppressor genes has been found in recent years,control growth, apoptosis and signal transduction of cell.The gene has two large Cp G island which is easy for methylation,once methylated,the RUNX3 gene will loss of expression,down-regulate target gene transcription and inhibiting cell proliferation which come about through the TGF-β and BMP signaling pathways will be restrained,then tumors occur.The present study indicating,the expression of RUNX3 gene is associated with the development of a variety of malignant tumors, especially the relationships with digestive tract malignant tumor has become a research hotspot in recent years. This experiment using immunohistochemical staining method,combined measurement the expression of D2-40 and RUNX3 in 76 colorectal cancer tissues and corresponding normal colorectal tissues,analysised of the relationship between them with colorectal cancer clinical pathological factors,and discussed primarily the correlation of the two. Methods: Collecting 76 cases colorectal specimens from colorectal cancer surgery patients, which were primary colorectal cancer and diagnosed for the first time from June 2014 to December 2014 in Gastrointestinal Surgery, Luzhou Medical College Hospital, all patients prior to surgery were not for radiotherapy, chemotherapy and biological treatment. While selecting the corresponding 76 cases normal tissue from surgical resection specimens. All cases have clinical data and pathological data.Immunohistochemistry method was used to detect the expression of D2-40 and RUNX3 in both colorectal cancer tissues and normal tissues, analysising their relationship between clinicopathological factors and with each other. Results: 1. D2-40 in colorectal cancer and normal colorectal tissue specimens were positive expression,the quantitative indexe LMVD in carcinoma peripheral area was significantly higher than central and normal colon tissue,the differences were statistically significant(P < 0.05);Central center and normal colorectal tissues LMVD differences had no statistical significance of the organization. 2. The LMVD of intra-tumoral area in colorectal carcinoma had nothing to do with patients’ gender, age, tumor size, invasion depth, degree of differentiation, tumor TNM stages, lymph node metastasis, and liver metastasis. The LMVD of peri-tumoral area in colorectal carcinoma was related to invasion depth,TNM stages, lymph node metastasis, and liver metastasis(P < 0.05);and,had nothing to do with patients’ gender, age, tumor size, degree of differentiation. 3. The positive expression rate of RUNX3 in colorectal cancer was 36.84%(28/76),and in normal colorectal tissue samples was 100%(76/76),compared with the positive expression of RUNX3 in cancer and normal colorectal tissues was significant differences(P<0.001). 4. The expression of RUNX3 in colorectal cancer was related to TNM stages, lymph node metastasis, and liver metastasis(P < 0.05), and had nothing to do with gender, age, tumor size, invasion depth,degree of differentiation.5.The LMVD of peri-tumoral area and the expression of RUNX3 in colorectal cancer was negative correlation(r=-0.478,P<0.05). Conclusion: 1.Lymphangiogenesis occurred in human colorectal carcer,and mainly distributed in cancer peripheral area.The increased LMVD in peri-tumoral area associated with the progression of colorectal cancer.2.The increasing LMVD in peri-tumoral area was related to invasion depth,TNM stages, lymph node metastasis, liver metastasis,and had nothing to do with patients’ gender, age, tumor size, degree of differentiation in colorectal cancer. 3.Low expression of RUNX3 associated with the development of colorectal cancer,the decreasing expression of RUNX3 was connected with TNM stages, lymph node metastasis, and liver metastasis obviously in colorectal cancer.RUNX3 is one of the poor prognosis of colorectal cancer biology index. 4.The LMVD of peri-tumoral area and the expression of RUNX3 was negative correlation in colorectal cancer,low expression of RUNX3 may promote lymphangiogenesis,and then promote lymph node metastasis. 5.Detecting the expression of RUNX3 and the LMVD of peri-tumoral can help to estimate progress, metastasis,and prognosis of colorectal cancer,regulating the expression of RUNX3 and anti-lymphangiogenesis may be the new target site for colorectal cancer molecular therapy.
