Effect Of 18β-Glycyrrhetinic Acid On Aquaporin 1,5 Expression In The Nasal Mucosa Of Rats With Allergic Rhinitis

Abstract/Summary:
Objective To investigate the influence and its significance of 18β-glycyrrhetinic acid on the expression of aquaporin 1,5 in rat nasal mucosa of allergic rhinitis.Method Sixty-four specific pathogen free rats were divided randomly into model group (MC group) and blank group (KB group).After AR model induced by dripping ovalbumin,the structure of rats nasal mucosa were observed by electron microscope in MC and KB group.Afterwards,the model rats were divided into three groups:allergic rhinitis group(AR group) group,loratadine group (as positive control drug,L group) and 18β-glycyrrhetiinic acid group(18β-GA group). Each group were intervened with different drugs severally. After 2 weeks,4 weeks,6 weeks,and 10 weeks,the expression of aquaporin 1,5 in each group were investigated and contrasted by immunohistochemical technique, and the change of the nasal mucosa tissue structure were investigated and contrasted by Hematoxylin-eosin staining.Result 〦lectron microscope:In MC group,the cilia of rat nasal mucosa columnar cell were lodging and sticky, goblet cells and gland cells contains a large number of secretory granules,the cytoplasm of capillary endothelial cell were thin and fracture. In KB group,rats nasal mucosa tissue structure is normal.②Hematoxylin-eosin staining:there were angiotelectasis, intercellular broadening and disordered arrangement of cells in nasal mucosal of AR group.However,there were no obvious pathological changes in rat nasal mucosa tissue of KB group.with the extension of drug delivery time, the pathological change in 18β-GA and L group nasal mucosa were improved and almost equivalent to KB group.③Immunohistochemical technique:The distribution of aquaporinl,5 in all groups were consistent. Aquaporin 1 expressed mainly in capillary and blood sinus endothelial cells,and connective tissue.Aquaporin 5 expressed intensively in Gland cells and ciliated columnar cells.④The statistical result of immunohistochemical technique suggested that the quantity of aquaporin 1,5 in AR group were markedly higher than KB group (P<0.05),and the expression of aquaporinl,5 in L group and 18β-GA group were gradually decreased and almost equivalent to KB group over time.Conclusion 18β-glycyrrhetinic acid may relieve symptoms and improve pathological changes of allergic rhinitis by reducing the expression of aquaporinl,5.
